Objectives: The response of the human periodontal ligament fibroblsts(HPDLFs) to mechanical stress are thought to play a crucial role in the remodling of periodontal tissues during orthodontic tooth movement. The purpose of this study was to analyse the effects of different static compressive stress on the proliferation and I type collagen synthesis of HPDLFs and to explore the mechanism of the remodling of the pressure side of the periodontal tissues during the Orthodontic treatment.Materials and Methods: the HPDLFs were isolated and cultured in vitro from healthy premolars of orthodontic patients, which were 10~14 years old. the cultured HPDLFs of the fourth passage were subjected to the different static compressive stress with a magnitud of 0KP, 15KP, 30KP, 45KP respectively for 1 hour. After that the proliferation was detected by flow cytometry , and the intracellular expressin of collagen type I was measured by immunocytochemical techniques.Results: Under the compression with a magnitude of 15KP, 30KP, the proliferation of HPDLFs greatly increased, and the percentage of the content of DNA of cells in S phase increased too(P<0. 05).But they dropped under the compression with a magnitude of 45KP;Besids this , it was found the intracellular expression of collage type I had a significant increase after... |
