Objective: To investigate the effects of osteoprotegerin (OPG)on the viability and IL-12 production of dendritic cells(DCs). So as to explore the effects of osteoprotegerin on immuno-inflammatory reactions in atherosclerosis.Methods: Mononulear cells were isolated from normal humanperipheral blood leukocytes, and cultured with the combination of rhGM-CSF,rhIL-4 and rhTNF-α to induce the mature phenotype of DCs. DCs were cultured in complete medium with receptor activator of NF-κB Ligand(RANKL) in the presence or absence of OPG . The cell survival was assessed for 4 successive days by trypan blue exclusion. The concentrations of IL-12 in the supernatant of DCs cultured with RANKL and OPG of different concentrations were determined by ELISA.
