Purpose:To construct the recombinant adenovirus of human endostatin (Ad-ASP-hES) ,and to infect the HRPE cells with it in vitro. The expression of hES in the HRPE cells was also observed after transfection. Laid the foundation for studying angiogenesis gene therapy by endostatin.Methods:1. Constructed and identified the Ad-ASP-hES by using the Adeasy 1 system methods.2. Normal HRPE cells were cultured in vitro ,and were identified by observing the morphous and immunocytochemical staining of keratin.3. Infected the HRPE cells with Ad-ASP-hES in vitro and observed the efficiency of the transfection by fluorescence microscope.
