Objective: To primarily evaluate whether there are the similar effect of a selective COX-2 inhibitor nimesulide on the growth of COX-2-negative and COX-2-postive human epithelial ovarian cancer(EOC) cell line xenograft tumor in nude mice, which would contribute to the further study whether we should assess the COX-2 profile of the EOC before initiating therapy with a selective COX-2 inhibitors. Methods: The EOC cell lines SKOV-3 and OVCAR-3 were cultured in RPMI 1640. Immunohistochemistry (with SP method) was utilized to measure the COX-2 profile of the cell lines. We use the EOC animal model to study the effect of nimesulide on the growth of the cell lines xenograft tumor. To measure the volume of tumors during the study, and then drawing a growth curve. We compared the difference of volume between the nimesulide-group and control-group when the experiments finished. The vascular endothelial growth factor (VEGF) levels in the tumors were measured by enzyme-linked immuno-adsordent assay (ELISA).
