Objiective to study the pancreatic β cell autophagy and possible relationship with islet oxidative stress.Materials and methods insulin resistance was induced in male wistar rats by feeding high-fat-diet which provided 66% calories as fat; at the same time the normal rats as control were fed with regular chew ,which supplies 10% calories as fat ,rats in both groups were sacrificed at setting 5 stages as 10,14,16,18,20weeks respectively, and serum insulin,blood lipids, fasting blood glucose , free fatty acids were measured at each time, HOMA-IR and ISI used to evaluate the degree of insulin resistance. the Rat islet β cells autophagosome were observed by electron microscope.at the same time , pancreas histology slice were incubated in PBS containing 0.05mM monodansylcadaverine (MDC). Laser scan confocal microscope was used to observe the islet fluorescence and islet fluorescence intensity was measured .SOD and MDA were measured to evaluate the oxidative stress status in separated islets.Findings 1 .at the ten weeks , the rats with high-fat-diet had much higher body weight, VF/W rational , insulin and lipid levels and HOMA-IR index increased ; insulin sensitivity index was decreased (1.35 ± 0.21 vs. 0.38 ± 0.02 p<0.05 ) correspondingly. all the above indicate the successful establishment of IR model. 2.SOD in HFD rat islets decreased at the fourteen weeks, and MDA increased 3. some visicoles were find in the cytoplasm at the 20 weeks .4. the fluorescence intensity of monodansylcadaverine was...
