[Objective] To investigate the effect and mechanisms of transplantation tolerance induced by "bone marrow cell transplantation + cycolp-hosphamide (cy)" protocol combined with short-course application of cyclosporine A.[Methods] Thirty-six male New Zealand rabbits and thirty-six female rabbits were used as donors and recipients respectively, matching pairs randomly. The thirty-six pair rabbits were divided into 6 groups randomly: blank control group, simple pre-treated group, simple BMT group, experimental group 1(CY+CSA+BMT), experimental group 2 (CY+CsA+fused bone marrow cell infusion) and third party donor control group.Skin grafting was performed on the same day of bone marrow transplantation. The number of bone marrow cells each was 3-4×10~8/kg, the survival time of skin graft, rejection against the third party skin and mixed lymphocyte reaction were observed. The polymerase chain reaction (PCR) was employed for the detection of SRY to comfirm the chimerism.[Results] The survival time of grafted skin was significantly prolonged in the group 4 (44.67 ±3.20) and group 5 (39.67 ±1.21) , as compared with the group 1(10.83± 0.75), group 2(16. 33±1. 21), group 3(11.33 ± 0.82) ,and group 6(14.83 ± 1.47), respectively(P<0.01).
