Objective This study was design to evaluate the role of connective tissue growth factor (CTGF) in scar formation of the filtering bleb that occurs after trabeculectomy by observing the expression of CTGF in the scarring process of the filtering bleb after surgery and the effect of exogenous CTGF on scar formation of the filtering bleb,and analyzing the correlation of the expression of CTGF and transformation growth factor-beta(TGF-β). Experiments were performed in a rabbit model.Methods Two experiments were performed: (1) Trabeculectomy was randomly performed on one of eyes of 28 healthy adult rabbits, the other eye was not performed the surgery and was used as the control. On days 1,3,5,7,10,14 and 21 after surgery they were killed in batch. Trabeculectomy was not performed on two other rabbits, who were killed before surgery. Tissue was harvested from the bleb area and was made pathological section, then was H-E stained and immunostained and observed under light microscope. Protein expression was quantitative analyzed by image analysis technique .(2) Trabeculectomy was performed on 12 eyes of 6 healthy adult rabbits, then these eyes were randomly divided into 3 groups and every group have 4 eyes. Exogenous CTGF and TGF-β and physiological saline was injected into filtering blebs, then intraocular pressure (IOP) was measured and filtering blebs were observed after surgery. The data was analyzed statistically by SPSS 12.0 for windows.Results (1)Histological examination showed that fibroblasts, inflammatory cells, collagen and neogenetic blood vessel occurred in the early stage after surgery, then increased over time and fibroblasts peaked by day 5 after surgery.
