Role Of Heat Shock Protein 70 Induced By Glutamine On Vascular Hyporeactivity Of Rats In Endotoxin-induced Septic Shock And Potential Mechanism

Part 1 Glutamine induces the expression of HSP70 mRNA in the rat Objective To observe the expression of HSP70 mRNA in the rat induced by glutamine. Methods 1) Twenty four healthy Sprague-Dawley rats were randomly divided into four groups (n=6 in each group): groupⅠreceived saline iv ;groupⅡreceived gln 0.15 g?kg-1 i.v; groupШreceived gln 0.45 g?kg-1 i.v.; groupⅣreceived gln 0.75 g?kg-1 i.v。All rats were killed after 4 h administration of Gln. HSP70 mRNA from heart,hepar,lung and aorta as well as GAPDH mRNA were measured by reverse transcription polymerase chain reaction (RT-PCR). 2) Thirty healthy Sprague-Dawley rats were randomly divided into five groups (n=6 in each group). All rats were killed at 1 h (group 1h),6 h (group 6h),12 h (group 12h),24 h (group 24h) and 48 h (group 48h) after administration of gln (0.75 g?kg-1 ). HSP70 mRNA from heart,hepar,lung and aorta as well as GAPDH mRNA were measured by reverse transcription polymerase chain reaction (RT-PCR).Results 1. The expression of HSP70 mRNA in groupⅡ,ШandⅣwere higher than groupⅠ(P<0.05), and the expression in groupⅡ,ШandⅣincreased gradually。2. The expression of HSP70 mRNA in group1h,6h and 12h was higher than in 24h and 48h (P<0.05)。Conclusion Glutamine could induce the expression of HSP70 mRNA in the rat,and it occurs the dose relation and the time process。...
