The roots of Indigofera spp have been used as main substitutes for traditional Chinese drug"ShanDouGen"in many provinces of China such as Shanxi, Henan, Hubei, Anhui and so on, usually called"Mu-Lan-Shan-Dou-Gen".In this paper, we studied the chemical constitutes of the roots of Indigofera kirilowii Maxim. which is one of the main resources of"Mu-Lan-Shan-Dou-Gen". Fifteen compounds were isolated and purified from the alcohol extracts by solvent extraction, silica gel column chromatography and re-crystallization. On the basis of various modern spectral analysis(IR,FAB-MS,1HNMR,13CNMR,DEPT,~1H-~1HCOSY,HSQC,HMBC)and classical chemical methods, the structures of 12 compounds were elucidated as 2-O-acryloyl-3,6-di-O-[3-nitropropanoyl]-α-D- glucopyranose (IKrC-1),corollin (IKrC-2), 2-O-acryloyl-3,4,6-tri-O-[3-nitropropan- oyl]-α-D-glucopyranose (IKrC-3), 3-O-acryloyl-2,6-di-O-[3-nitropropanoyl]-α-D- glucopyranose (IKrC-4),β–sitosterol (IKrC-5), lupeol (IKrC-6), 3-nitropropionic acid (IKrC-8), 4-O-acryloyl-2,3, 6-tri-O-[3-nitropropanoyl]-α-D-glucopyranose (IKrC-9), betulinic acid (IKrC-10), daucosterol (IKrC-11a), stigmasterol–3-O– glucopyranoside(IKrC-11b), 2,3,4,6-tetra-O-[3-nitropropanoyl]-α-D-glcopyranose (IKrC-12). Among these compounds, IKrC-3 (kirilowin A), IKrC-1 (kirilowin B), IKrC-4, and IKrC-9 were new compounds, and the other compounds were reported from I. kirilowii for the first time.We also summarized the chemical and bioactive studies on Shan- Dou-Gen and Indigofera spp. Meanwhile, the structures, NMR spectroscopic characteristics and physico-chemical properties of 3-nitropropanoyl glucopyranoses were summarized.
