(2) The appropriate corneal acellular extracellular matrixes were studied and the holes on the biomaterials were made. The growth of corneal cells on the corneal acellular extracellular matrixes and biomaterials was researched. (3) The effect of laminar corneal transplantation with corneal acellular extracellular matrix of high performance was preliminarily investigated.Method: (1)The rabbit corneal epithelial cells, keratocytes and endothelial cells were cultured by using tissue inoculation or digestion methods and the cells were passed on after they become confluent and the growing characteristics were observed every day. (2) To select the appropriate method to make corneal acellular extracelluar matrix basing on the several ways and compare the biological characteristic with nature cornea. (3) The procedure with frozen-drying and chemistry were used to produce porous structure in the biomaterials. (4)Rabbit corneal epithelial cells, keratocytes and endothelial cells were cultured on corneal acellular extracelluar corneal matrix and biomaterials. The growth, arrangement and adhesion of cultured cells were observed. (5) The corneal acellular extracellular matrix of high performance was transplanted on the rabbit corneal and the effect of post-operation was observed.Result: (1) The epithelial cells, keratocytes and endothelial cells adhered and grew well on the culture flask and plates. The confluent cell monolayer could be formed. The shapes of cells were typical and the subculture cells still maintained the characteristics of the primary cells. (2) The corneal acellular extracelluar matrix processed by trypsinization-frozen method showed better results, which not only made a relative thorough acellular matrix but also maintained the structure of nature corneal matrix.
