Objective:People's quality of life is severely affected by myocardial ischemia. Myocardial ischemia is one of diseases with high morbidity and high mortality. Its prevention and cure have been become an emphasis in the medicine field. Xinmaitong is a compound recipe of traditional Chinese drug to prevent and cure ischemic heart diseases. Its main functions include enhancing blood circulation and removing stasis, and relieving pain. Clinical practices and experimental studies have shown that Xinmaitong can prevent injury of myocardial ischemia. To probe its cellular and molecular mechanism of Xinmaitong against the myocardial ischemia injury, we evaluated protective effects of Xinmaitong on impaired CMCs induced by H2O2 and on the ischemic heart disease (IHD) induced by ISO in rats, and observed changes of TGF-β1, TβRI,TBRII, and NF-κB expression during myocardial ischemia.Methods:1.The protective effects of Xinmaitong on the H2O2-induced CMC's injury: Preparation of CMCs of neonate rats was done by the enzyme digestion and adherence of unequal velocity. CMCs were randomly divided into three groups: Control group (15% non-medicated serum+DMEM), model group (15% non-medicated+H2O2+DMEM), and medicated serum group (15% Xinmaitong-medicated serum +H2O2+DMEM). Cells were seeded in 96 or 24 well culture plates, and were cultured in 37°C, 5% CO2, and 95% humiditied condition. After 24h, CMCs were treated by H2O2 (including 0, 50, 100, 200,and 400μM final concentration groups). CMCs activity at 8h was detected with MTT. The impaired degree of CMCs at Oh, 0.5h, 1h, 2h, 4h, 8h (200μmol/L H2O2 final concentration) was analysed with LDH assay kit and MDA assay kit.2.
