| The research of anti-tumor active substances from microalgae, which were rich in the sea, becomes a new hotspot. A model of screening cytotoxic substances was established in the thesis. It weeded out 10 species microalgae which contains cytotoxic activity to the Hela cell line: Isochrysis galbana Parke H29, Isochrysis galbana Parke 3011, Isochrysis galbana Parke 8701, Platymonas subcordiformis, Chlorella saccharophila, Chroomonas saccharophila,Phaeodactylum tricormutum, Chaetocoreros muelleri, Dunaliella, Anabaena, Scenedesmus.The results showed that there were seven organic extraction of Isochrysis galbana Parke H29 had strongly inhibited the growth of Hela cells. The rates of inhibition were 90.32 %, 93.79 %, 90.16 %, 91.23 %, 95.91 %, 96.45 % and 90.83 %, which were from the different extrction like Cyclohexane, Ethyl acetate, Aether, Methyl benzene, 1-amyl alcohol , Acetone and Ethanol, respectively.During the experiment of optimizing the Isochrysis galbana Parke H29, it had been found that its optimum growth temperature is 24℃, and light intensity is 4500 lux. Based on the F/2 medium, it was showed: the best N, Fe, P resources were NaNO3, FeC6H5O7·5H2O and KH2PO4. Acetone extracts separated from Isochrysis galbana Parke H29 by using TLC and HPLC, two active substances were obstained after determinations of the nature and purity, each substance has highly purity and might be one Anthraquinone.Throughout the research, Anti-tumour active substances from Isochrysis galbana Parke H29 was found for the first time. It most possibly could find new anti-tumor lead compounds, so further research of Isochrysis galbana Parke H29 could be investigated . |
