Objective :To investigate the effects of mifepristone and gestrinone on growth,proliferation and apoptosis, in isolated ectopic endometrial stroma cells and normal endometrial stroma cells in vitro.Methods :Isolated the axenic ectopic endometriotic tissues and normal endometrium tissues to culture in vitro. Then the cultured endometrial stroma cells were identified by vimentin immunofluorescence staining.. Endometrial stroma cells were cultured and exposed to mifepristone or gestrinone of different doses respectively. To observe the corpuscular growth condition by inverted microscope.The inhibition of the cells during 60 hours was determined by methylthiazolyl tetrazolium (MTT) assay, and the cell growth curve was made. Mifepristone or gestrinone was administered to the ectopic endometrial stroma cells and normal endometrial stroma cells and at 36 hours the cell cycle and apoptosis rate were monitored by flow cytometry ( FCM).Results :The purity of cultured ectopic endometrial cells and normal endometrial stroma cells was 95% identified by vimentin immunofluorescence staining. Mifepristone and gestrinone at different concentrations could inhibit the growth and proliferation of endometrial stroma cells in a dose-and time-dependent manner. FCM showed that after the exposure to different concentrations mifepristone or gestrinone for 36 hours, the cell cycle were inhibited from G1 stage to S stage,the number of cells at G1 stage increased obviously.The apoptotic rate of endometrial stroma cells exposured to different concentrations also increased.But the inhibition of the normal endometrial stroma cells is poorer than it of the ectopic endometrial stroma cells.Conclusion: 1 .Mifepristone and gestrinone can also significantly inhibit the growth and proliferation and induce the apoptosis of ectopic endometrial stroma cells.To compare the effects of mifepristone and gestrinone at the same concentration ,the latter was more strong. 2.The inhibition of mifepristone and gestrinone to the ectopic endometrial stroma cells was stronger than to the normal endometrial stroma cells.
