| Part I The relationship between the expression of MMP-9 in sinonasal inverted papilloma and its recurrence andmalignancyBackground and ObjectiveSinonasal inverted papilloma(SNIP) is a benign tumor that is characterized by aggressive nature and high recurrence rate and malignant transformation potential. The etiology of the lesion and the mechanism of its correlation to malignancy and recurrence are still not well understood. The MMP-9 is a kind of highly conserved metalatom-dependent endopeptidases, which is capable of degradation of most components of the basement membrane and extracellular matrix. MMP family has been studied in most tumour types. Overexpression of MMPs is now known to be a characteristic of most malignant tumours, especially in tumor infiltration and metastasis. The objective of this study is to detect the expression of MMP-9 and to investigate the biological significance in its recurrence tendency and malignant potential.MethodsForty specimens with SNIP and ten specimens with squamous cell carcinoma (SCC) were selected in the experiment. All the specimens were sorted into four groups, which were SNIP, recurrent SNIP, SNIP with carcinoma, and squamous cell carcinoma (SCC). The expressions of MMP-9 were analyzed by using immunohistochemistry and the results were analyzed according to thequantitative immunohistochemical analysis.ResultsSignificant increased expressions of MMP-9 was observed in SNIP, recurrent SNIP, SNIP with carcinoma, and SCC.According to the rank test, the prominent differentiation in the expression of MMP-9 was observed between SNIP and recurrent SNIP, SNIP and SNIP with carcinoma, SNIP and SCC. But between recurrent SNIP and SNIP with carcinoma, recurrent SNIP and SCC, SNIP with carcinoma and SCC, there was no prominent differentiation. MMP-9 was mostly expressed in the basic layer in SNIP and recurrent SNIP,and its expression was strengthened in the almost whole epithelial layer and the cell interstitial substance. ConclusionThe expressions of MMP-9 was involved in the mechanism of SNIP occurrence and played an important role in its recurrence and malignancy. The over expression of MMP-9 may be associated with early events in the recurrence tendency and carcinogenesis / transformation to SNIP.Part II The relationship between the expression of PCNA in sinonasal inverted papilloma and its recurrence andmalignancyBackground and ObjectivePCNA which was called proliferating cell nuclear antigen,was a kind of acidity nonhistone protein nucleo-albumose polypeptide,it was play a important role in the reproduction of DNA,and could be regarded as the specificity mark of the s stage, the biology behavior of the tumor was interrelated closely with the activity of proliferation.This interview aimed To explore the function of PCNA in the occurrence, recurrence and malignancy of the sinonasal inverted papilloma (SNIP),to direct the clinic to observe the patients in high danger,and advanced the level of clinic diagnose and cure. MethodsForty specimens with SNIP and ten specimens with squamous cell carcinoma (SCC) were selected in the experiment. All the specimens were sorted into four groups, which were SNIP, recurrent SNIP, SNIP with carcinoma, and invasive squamous cell carcinoma (SCC). The expressions of PCNA were analyzed by using immunohistochemistry and the results were analyzed according to the quantitative immunohistochemical analysis. ResultsSignificant increased expressions of PCNA were observed in SNIP, recurrent SNIP, SNIP with carcinoma, and SCC. According to the rank test, in the expression of PCNA, there was no prominent differentiation in every two groups. The PCNA-positive cells always weekly expressed in the basic and middle layers within the two-thirds level in SNIP; The positive expression was strengthened in the recurrent SNIP;Moreover, the expression of positive cells gradually increased and extended to the entire epithelial layer in SNIP with carcinoma and SCC, the poorly differentiated expressed unorderedly. ConclusionThe expressions of PCNA was involved in the mechanism of SNIP occurrence and played an important role in its recurrence and malignancy. The over expression of PCNA may be associated with early events in the recurrence tendency and carcinogenesis / transformation to SNIP. |
