Separation And Culture Of Rabbit Vaginal Smooth Muscle Cells In Vitro

Objective:To research the cultural method of vaginal smooth muscle cells in vitro. It is in order to provide the seed cells for vaginal tissue engineering and prepare for the virginal SMC research in vitro later. Method:(1) Separated the vaginal smooth muscle tissues from New Zealand female rabbits and then cultured vaginal smooth muscle cells by tissue explant and tissue explant + collagen digestion methods respectively. Observed and then write down the primary cultural time of each method. Serial subculture is used to gain the purer vaginal SMC.(2) Monitored the cell morphology and its statue of growth and proliferation under the inverted microscope.(3) Survey the cell growth by MTT experiment.(4) Observed the surface structures of the vaginal SMC under scanning electron microscope .(5) Observed the ultramicrostructure of the vaginal SMC under the transmission electron microscope.(6) Detected the expression of SM a -actin by immunohistochemical methods.Result:(1) The vaginal SMC which were obtained by the methods mentioned above could propagate for 5-6 generations.
