| Part one: The expression of glutamine transporters in guinea pig cochlearObjective :To detect the expression of ASCT2, LAT1 , LAT2 ,SN1,ATA1 and ATA2 in guinea pig cochlear and explore their influence on the mechanism which glutamine in supporting cells were transported into inner hair cells during glutamate-glutamine cycle. And the study would possibly offer the foundation of research in molecular biology on the glutamate-glutamine cycle.Methods :The total RNA of guinea pig cochlear was extracted by Trizol one step method and the expression of ASCT2,LAT1 ,LAT2 ,SN1,ATA1 and ATA2 in cochlear was detected with reverse transcription polymerase chain reaction.Results : ASCT2,LAT1,SN1 and ATA1 ,were expressed in guinea pig cochlear ,and ASCT2 was the strongest,while there were no expression of LAT2 and ATA2 in guinea pig cochlear.Conclusion :The presence of glutamine transporter families in guinea pig cochlear suggested that glutamine in supporting cells was transported into inner hair cells might through glutamine transporters. and this experimental result supported the hypothesis on existence of glutamate-glutamine cycle in cochlea. |
