Attention deficit hyperactivity disorder (ADHD) is an early-onset, clinically heterogeneous disorder of inattention, hyperactivity and impulsivity, often companying with study difficulty, lowly self-confident, conduct obstacle etc. At present, the cause of ADHD is unknown, but it is thought to be biological, psychological and social. This research wants to look for the clues of this disease only from the angle of the neuro-biochemistry and to explore the possible pathophysiological mechanism in ADHD.The content of monoamine neurotransmitters and amino acids inside the body is very low, so to develop a sensitive and accurate method to determine those neurotransmitters is the basic of studying the bio-chemical mechanism of ADHD. High performance liquid chromatography for simultaneous separation and rapid determination of amino acid and monoamine neurotransmitters were developed.Several main parts of this paper were summarized as follows:1. Development and evaluation the method for determination of amino acid neurotransmitters by HPLC-FluA high performance liquid chromatography for simultaneous separation and rapid determination of five amino acid neurotransmitters [glutamate(Glu), aspartate(Asp),γ-aminobutyric acid( GABA ), glycine(Gly), taurine(Tau)] in the striatum of rat brain was developed. The HPLC column was Kromasil C18 (4.6×250 mm, particle size 5μm). The precolumn derivatization reagent was 1-dimethyl aminonaphthalenesulfonyl chloide and trazodone was used as internal standard. The mobile phase was a mixture of 42% methanol and 58% 0.1mol/L acetate sodium (v/v), which involved 1% tetrahydrofuran, 14mmol/L1-Heptanesulfonic Acid, Sodium Salt. Isocratic elution was used.
