PURPOSE: The objective of this study was to determine the expression of HER-2, COX-2, CD105, Laminin-R and Paxillin in breast carcinomas using tissue microarray techonolgy(TMA), and investigate their clinical and prognostic valuation.METHODS: 192 paraffin blocks of pathological confirmed breast invasive ductal carcinomas were selected from hospitalized patients during 1995 to 2000. Marked the target region on the HE slice under low power lens, and made a sign on the corresponded paraffin block. These paraffin blocks were called donator blocks. Then made hard-fitted receptor block(50mm×30mm×20mm). Extract tissue core of 1.2mm in diameter from those 192 donator blocks, 7 definited positive paraffin blocks (ER, PR,HER-2,COX-2,Paxillin,CD105,Laminin-R,CD105) and 7 definited negative paraffin blocks (ER, PR, HER-2, COX-2, Paxillin, CD105, Laminin-R, CD105) respectively. Insert the cores into the receptor block and then two tissue microarrays were completed. Continuously sliced for 4 μ m and stained by HE, then eliminated the samples of losing, wrong tissues and insufficiency tissues.
