Objective:On the basis of ovariectomied animal model, we investigate the proliferation capacity and differential capacity to osteoblast and adipocyte of MSCs, and the proliferation and antioxidant effect of naringin on MSCs in ovariectomied rats .The study may provide a better understanding of the mechanisms involved in the pathogenesis of postmenopausal OP, and another aim of this study is to illuminate the biological property of Drynaria fortunei on OP.Methods:1. The animal model of osteoporosis was established by ovariectomied method in 10-month -old female Sprague-Dawley rats (SD).2. MSCs were isolated from the rats by means of the density-gradient centrifugation and cell attachment to tissue culture plates 3 months after operation. The cell surface antigens were detected by flow cytometry (FCM) for identification of the MSCs.3. The growth curve, adhesion rate was investigated to illuminate the proliferative potential of MSCs derived from ovariectomied osteoporotic rats. Capacity of MSCs differentiate was evaluate down the osteoblst by investigating the differential ratio of these MSCs to osteoblast and adipocyte.4. MTT method was used to investigate the proliferation effects naringin on MSCs in ovariectomide rats, and to determine the optimal concentration of naringin. Cell cycle and proliferation index (PI) of MSCs was detected to demonstrate the effects of naringin on proliferation of MSCs.5. ALP staining was used to study the effects of naringin on osteogenic differentiation of MSCs.6. The third passage MSCs were divided into control group, induced group and naringin group, to illuminate the effects of naringin on osteogenic differentiation of MSCs.
