| Objective: Mongolian Traditional Medicine (MTM) has a long history. It not only has a unique theory but also a striking national feature. For thousands of years, it has made a great contribution to the health of mankind. Particularly in modern time, to return to nature and attach importance to traditional medicine has become a significant trend in the world. Traditional medicine has attracted global concern and preference. In recent years, many researches concerned in mechanism of promoting animal immunity with MTM have made an encouraging progress. Some of MTM has a various effects on immunologic functions. These researches shows that MTM has effects on promoting the immune organs and developing the immunity of animal organisms, and increased animal resistance to disease by activating monocytemacrophage cell and natural killer cell (NK cell). MTM also has effects on promoting the hyperplasia of lymphocyte B and T as well as to produce immunoglobulin (Ig) and also some evidences shows that MTM has a potential function in preventing and treating of tumor and therefore it is an important issue needed for further study. The more understanding of the immune mechanism of MTM, the more ideal and effective MTMs can be found in the future. It will be used in immunologic diseases and anti-tumors etc. Mongolia Traditional Medicine Preparation San Hua San (SHS) derives from the classical works of MTM. It has the functions of relieving internal heat or fever and a subsidence of swelling. As a result, it is used for such diseases as plague, fever syndrome, scabies. After years of medical experiments, it is functioned as decreasing inflammations, resisting virus, and preventing human body from tumors. It is predicted that SHS may regulate human immune system. But so far there are not many researches on its mechanism. The purpose of the test is to find the effects of Mongolia Traditional Medicine PreparationSHS on immune level in mice and probe into the effects of SHS on immune mechanism system so as to offer scientific basis for clinic application of Mongolia Traditional Medicine Preparation SHS, realize the modernization of national medicine and make contributions to the cause of human health.Methods: This experiment uses SHS as the experimental medicine, and mice as the experimental animals. It chooses pure Balb/c mice, the weight of which are 20±2 g. Half of them are male, and half are female, with standard stuff and fresh water to feed them. The mice are divided into 4 main groups, each having 40 mice. Each main group is subdivided into 4 small groups and 10 mice in each small group. The 4 small groups are: negative contrast group (physiological saline 0.2ml/q.d for each mice), positive contrast group (cyclophosphamidum 0.05g/kg.qd),SHS experimental group 1 and SHS experimental group 2. The animals in positive contrast group were examined by appropriate mode. Mongolia Traditional Medicine Preparation SHS of different doses were fed to mice continuously in force (mice in SHS experimental group 1 were given containing 0.75g/kg.q.d and SHS experimental group 2 were 1.5g/kg.q.d of Mongolia Traditional Medicine Preparation SHS). After several days, the samples in each group were collected for testing. It demonstrated through the total white blood cell count and white cell classification amount, the transformation mice of lymphocytes, the phagocytes percentage and index of peritoneal macrophage that the effect of Mongolia Traditional Medicine Preparation SHS on the mice. Moreover, the two indexes about the lymphocytes'activation by RT-PCR, such as perforin and Fasl were observed. Method of extracting SHS liquid: take raw medicine 100 g, then put 250 ml purified water, and stew them until the liquid reaches 200ml. After that, filter the medicine to get the strong liquid with the help of alcohol. The extracted liquid will be 50 ml, and final liquid product will be 200g/ml for medical use.Results: the results showed as follows: group 1 and 2 could improve the total white blood cell count and white cell classification amount and could significantly increase the transformation mice of lymphocytes compared to the contrast group (P<0.01); it showed that SHS could significantly increase cellular immune function of mice; SHS could significantly enhance the phagocytic activity of peritoneal macrophage compared to the contrast group (P<0.01); it also showed that SHS could promote the nonspecific immune function of mice; These were enhanced significantly when the immunology function of mice was inhibited by Cy (P<0.01); it showed that the SHS enhanced the killing activity of the lymphocytes obviously-exhibiting as the expressionof the two (Fasl and perforin) index increasing obviously.Conclusions: These experiments indicated that Mongolia Traditional Medicine Preparation SHS was able to activate immune function of mice and enhance Fasl and perforin to express in splenetic.These tests will provide sufficient scientific basis for clinic application of Mongolia Traditional Medicine Preparation SHS... |
