Hepatocellular carcinoma (HCC) is a common malignancy worldwide and is a leading cause of death. Metastasis and recurrence is the major cause for lower post-operation survival rate of HCC patients. Like other cancers, the development and metastasis of HCC is a multifactorial and multistage pathogenesis. But mechanism of metastasis is not clear now. Lymphatic metastasis is regard as a early signal of metastasis, so research in lymphatic metastasis mechanism can help to early diagnosis of tumor and effective therapy.Two-dimensional electrophoresis and mass spectrometry was used to display and compare the differential expression of lymphatic metastasis related proteins in Hca-F and Hca-P cells and in order to make the foundation for further studying lymphatic metastasis mechanism. The results suggest that one of differential expression proteins, ERp29, were higher in Hca-P than in Hca-F. Because of obvious expressing difference of ERp29 between Hca-F and Hca-P, we chose this molecule as the target for following study. By western blot, immunocytochemistry, immunofluorescence and flow cytometry analysis on protein and cell level, we validate expressing level of ERp29 in Hca-F and Hca-P.Methods: In this study, western blot, immunocytochemistry, immunofluore- scence and flow cytometry analysis were used to detect the expression of ERp29 in Hca-F and Hca-P.Results : The results of immunocytochemistry, immunofluorescencesuggest that ERp29 protein was located at cytoplasm of hepatic cells and some were also detected in the nucleus. Flow cytometry analysis suggest that positive expression ERp29 protein rates were 29.36% in Hca-F,45% in Hca-P. There were obvious expressing difference of ERp29 protein and this difference were statically﹙ P< 0.05﹚ . The results of western blot suggest that ERp29 had positive expression in Hca-F and Hca-P. Its expression in Hca-P were apparently higher than in Hca-F. And there were statically different between Hca-F and Hca-P﹙P<0.05﹚.Conclusion:1 ERp29 protein had positive expression in Hca-F and Hca-P. Its expression in Hca-P was obviously higher than in Hca-F.2 ERp29 protein was located at cytoplasm of hepatic cells and some were also detected in the nucleus.3 The expression of ERp29 protein in HCC had relationship with its pathogenesis and metastasis. It may become a new marker for the diagnosis of HCC and a target for prognosis and biotherapy.
