Objective: To investigate the frequency of HLA-DRB and -DQB1 alleles in the population of Han nationality in north China and susceptible gene of Eales disease in HLA-DRB, HLA-DQB1 Alleles.To analyze the relationship between susceptible gene and bilateral affection and visual acuity of patients. to establish the foundation of researches on diagnosis, treatment and prognosis of Eales disease in gene levelMethods: The subjects were analyzed by case-control study consisted of two groups such as normal control group and Eales disease group. DNA samples from 30 healthy people and 27 patients with Ealesdisease were detected by PCR-microSSP. The Alleles of HLA-DRB and -DQB1 from control group were compared with those from Eales disease group by SPSS12.0.The relationship between susceptible gene and bilateral affection and visual acuity of patients was further analyzed.Results: 15 HLA-DRB alleles and 7 HLA-DQB1 alleles in two groups were found out, and the incidence of HLA-DRB alleles between two groups had significant difference ,but that of HLA-DQB1 alleles hadn't difference.The frequency of DRB1*04 in patient group and control group was respectively 44.4% and 20.0%, P=0.047, OR=3.20, and there was a positive correlation between Eales disease and DRB1*04. The incidence of bilateral affection between groups with HLA- DRB1*04 -positive patients and HLA- DRB1*04-negative patients was separately 58.3% and 6.7%, P=0.008, OR=19.6, and there was also a positive correlation between incidence of bilateral affection and HLA- DRB1*04,but Comparison of visual acuity of patients between two groups hadn't difference. Conclusion: (1) HLA-DRB and -DQB1 alleles were typed by PCR-microSSP with speedness, accuracy and credibility. (2) The distributive characteristic ofHLA-DRB, HLA-DQB1 alleles in normal population and patients with Eales disease in the North Hans was obtained. (3) The results showed that HLA-DRB1*04 might be a susceptible gene of Eales disease .(4) There was a positive correlation between incidence of bilateral affection and HLA-DRB1*04.
