In the present paper a method for separation and determination of purines is described. It is divided into two main parts:The first part is simultaneous determination of adenine, hypoxanthine, xanthine and guanine by artificial neural networks-ultraviolet spectrophotometry without separation. The determination of the number of hidden nodes and learning rate, the selection of learning sample and the selection of pH of system have been investigated.The second part is determination of adenine, hypoxanthine and xanthine by HPLC. The effects of several factors on extraction rate in ultrasonic extraction, e.g.: volume of water and time of extraction have been investigated. The optimum chromatographic separation of mobile phase and its proportion, column temperature and flow-rate has been determined in the determination of HPLC. And purine compounds in zaocys, ground beetle, bufonid and lumbricus have been determined by the method.
