Study Differerce Of Antitumor Mechanism Of Tian Li Ke And Zedoary Turmeric Oil On HepG-2 Cell

Objective: To explore the effect of antitumor molecular mechanisms of Tian Li Ke (TLK) and zedoary turmeric oil (ZTO) and 5-Fu in HepG-2 .Methods: By cells culturing in vitro , anti-proliferation effects of ZTO and TLK were assessed with methyl thiazolyl tetrazolium (MTT) colorimetric assay . After treatment with drug, morphology of cells was observed under light microscope, fluorescence microscope and transmission electron microscopy respectively .The flow cytometry was used to detect apoptotic rates and cell cycle distribution .The expressions of telomerase were determined by the kaijiTRAP-argentation method . The expressions of survivin and NF-κB protein were determined by the immunocytochemical method.Results: MTT showed that anti-proliferation of ZTO on cancer cells of the liver,the inhibitory effects were obvious ,in a concentration-time dependent manner , neither TLK. Microscope showed that cell presented some morphologic features of apoptosis, including cell shrinkage, plasma membrane blebbing, nuclear condensation and formation of apoptotic bodies after expose to ZTO. A lot of vacuoles appeared in HepG-2 cytoplasm after treated with TLK. The flow cytometry showed that after exposure to drugs , cell-cycle was blocked at S phase . Tian Li Ke significantly changed cell cycle-arrested in S phase (P<0.01) and phosphatdylserine moved out of cell membrane after exposure to AV/PI. ZTO and 5-Fu can induce cell apoptosis. ZTO can induce cell death(P<0.01). After application of Tian Li Ke and zedoary turmeric oil and 5-Fu, expression of survivin protein,NF-κBp65 and telomerase decreased (P <0.05). Expression of survivin and telomerase protein decrease markedly after cells exposure to TLK (P<0.01) .Conclusion: TLK mainly anti-proliferate and make cell-cycle blocked at S phase . zedoary turmeric oil and 5-Fu can induce apoptosis and make cell-cycle blocked at S phase. TLK significantly changed cell cycle-arrested in S phase. TLK can induce cell early apoptosis .The possible mechanism is to reduce survivin protein,NF-κBp65 and express of telomerase.
