Apoptosis plays an important role in oncogenesis and tumor progression. For the purpose of studying the role of survivin in generation and progression of ameloblastoma and odontogenic keratocyst, we detected the expressions of survivin mRNA and protein in ameloblastoma and odontogenic keratocyst using RT-PCR and immunohistochemical assay respectively, and analyzed the correlation with the expressions of bcl-2, p53 and caspase-3.Partâ… Expression of survivin and its significance in ameloblastomasObjective: To study the expression of survivin in ameloblastoma, and the correlation with the expressions of bcl-2, p53 and caspase-3, to approach the possible role of them in oncogenesis and development of ameloblastoma.Methods: TUNEL was used to detect the apoptotic cell of ameloblastoma. RT-PCR was used to detect the expression of survivin mRNA in 5 cases of ameloblastoma. Immunohistochemical assay was used to detect the expressions of survivin, bcl-2, p53, caspase-3 protein in 21cases of ameloblastoma (including 11cases of primary ameloblastoma and 10 cases of recurrent ameloblastoma). Results: Apoptotic cells detected by TUNEL method were mainly seen in the central polyhedral cells of the ameloblastoma tumor nests. The expression of survivin was at both mRNA and protein levels in ameloblastoma. The expressions of survivin, bcl-2, p53, caspase-3 were 13(61.9%), 14(66.7%), 14(66.7%), 10(47.6%) cases of ameloblastoma respectively. Survivin, bcl-2 and p53 positive staining was mainly in the peripheral columnar or cuboidal cells of the tumor nests, caspase-3 was mainly in central polyhedral cells. There was a positive correlation between the expression of survivin and the expression of bcl-2 and p53, there was a different expression position between survivin and caspase-3.Conclusion: Survivin, bcl-2, p53, caspase-3 played an important role in oncogenesis and progression of ameloblastoma, and survivin may be a diagnosis marker and a potential therapeutic target for ameloblastoma.Partâ…¡Expression of survivin, bcl-2, p53, caspase-3 and their significance in odontogenic keratocystObjective: To investigate the expression of survivin, bcl-2, p53 and caspase-3 in human odontogenic keratocyst (OKC) and the possible role of them in the cystogenesis and development of OKC.Methods: TUNEL was used to detect the apoptotic cell of OKC. RT-PCR was used to detect the expression of survivin mRNA in 5 cases of OKC. Immunohistochemical assay was used to detect the expression of survivin, bcl-2, p53 and caspase-3 protein in 20 cases of OKC ( including 10 cases of primary OKC and 10 cases of recurrent OKC), and in 10 cases of dentigerous cyst, 10 cases of radicular cyst.Results: Apoptotic cells detected by TUNEL method were mainly seen in the flattened cells of surface layer of OKC. The expression of survivin was at both mRNA and protein levels in OKC. The expression of survivin, bcl-2 and caspase-3 were 13 (65%), 13 (65%), 9 (45%) cases of OKC respectively, the expression of p53 was negative in all cases. Survivin and bcl-2 positive staining were mainly in the columnar or cuboidal cells of basal layer. There was significantly higher positive ratio of OKC than dentigerous cyst and radicular cyst. Caspase-3 positive staining was mainly in surface layer flattened cells of OKC. The expression of survivin correlated with bcl-2 expression, there was a different expression position between survivin and caspase-3.Conclusion: Survivin, bcl-2 and caspase-3 played an important role in generation and development of OKC, the expression of survivin can be useful to differentiate OKC from other types of odontogenic cysts.
