| Objective : Growth differentiation factor-5 (GDF-5) have been suggested as pivotal signaling molecules in the developing skeleton. But if it to be involved in developing temporomandibular joint remains unknown: To study the expression of growth differentiation factor-5 in the mandibular condylar cartilage of developing temporomandibular joint of rat fetuses. Method : A series fetuses rat TMJ developing model was built histologically. Expression of growth differentiation factor-5(GDF-5) protein were observed by S-P immunohistochemical staining and western blotting in the mandibular condylar cartilage of developing temporomandibular joint of SD rats during prenatal at 13,15,17,19,21 days of gestation and postnatal l-5day. Results: GDF-5 was expressed in all observed groups and throughout the mandibular condylar cartilage in development of different periods, and the highest expression of GDF-5 was in chondroblastic cell layer and prolifetation cell layer.The expression of GDF-5 was low in 15 day of gestation,increase with age,then decrease in 21 day of gestation.WB result is that the expression of GDF5 increase gradually at postnatal 1-5 days. Conclusions: There were different expressions of GDF-5 in the condylar cartilage of SD rats developing temporomandibular joint . GDF-5 might be involved in the modulation of the development, regeneration and structure maintenance of chondrocyte. |
