Objective: To study the expression of matrix metallopreteinases (MMPs) andtissue inhibitors of metallopreteinases (TIMPs) in human gestational trophoblasticdisease and their predictive value in malignant transformation.Methods: The protein expression of MMPs and TIMPs was detected byimmunohistochemistry in 18 cases of normal villi,65 cases of gestationaltrophoblastic disease, (49 cases of hydatidiform mole,2 malignant transformed mole;6cases of invasive mole and 2 cases of choriocarcinoma).The mRNA expression ofMMPs and TIMPs was detected by RT-PCR in 18 cases of normal villi,35cases ofhydatidiform mole,in which 6 cases were malignant transformed mole..Result:1,The protein and mRNA expression of MMP-7 was detected in gesationaltrophoblastic disease.2,The protein expression of MMP-7,MMP-9 and TIMP-2 was significantlyhigher in malignant transformed mole,invasive mole and choriocarcinoma than thatin normal villi and nonmalignant transformed mole(p<0.05). The expression ofTIMP-1 in all cases has no significant difference (P>0.05).3,The mRNA expression of MMP-7,MMP-9, TIMP-1,TIMP-2 has no differencebetween normal villi, hydatidiform mole and malignant transformed mole (p>0.05).4,The expression of MMP-7 was positively associated with MMP-9(P<0.05),Theexpression of MMPs was positively associated with TIMPs(p<0.05).5,The ratio of MMP-9/TIMP-1 was significantly higher in malignant transformedmole,invasive mole and choriocarcinoma than that in normal villi and nonmalignanttransformed mole(p<0.05).6,The protein expression of MMP-7 and MMP-9 was significantly higher inhydatidiform mole with malignant high risk facters than that without malignant highrisk facters(p<0.05). The protein expression of TIMP-1 and TIMP-2 in all cases hasno significant difference (P>0.05). Conclusion:1,The expression of MMP-7 was detected in gesational trophoblastic disease. Itmaybe associated with blood vessel metastasis.2,Upregulation of the MMP-7,MMP-9,TIMP-2 with the malignancy degree in thegestational trophoblastic disease,But The expression of TIMP-1 in all cases has nosignificant difference.It indicates that MMP-7, MMP-9, TIMP-2 can reflect themalignancy degree of the disease to some extent.3,The high ratio of MMP-9/TIMP-1 is associated with the malignany and invasionof the gestational trophoblastic disease,It could be an useful index for prediction ofgestational trophoblastic diserse and provide the evidence to guide clinical chemicalthearpy.4,The expression level of MMP-7 and MMP-9 is high in hydatidiform mole withmalignant high risk facters,but there is no change for the expression level of TIMP-1and TIMP-2.It can disturb the balance of MMPs and TIMPs,witch induce tomalignancy.
