| Objective To evaluate the levels of serum IL-12, IFN-γand LDH in patients withdifferent types of acute leukemia (AL), including acute myeloid leukemia (AML) and acutelymphocytic leukemia (ALL).Methods The levels of serum IL-12 and IFN-γwere detected by ELISA and the level ofLDH was assayed by auto biochemistry analyzer. 20 cases of normal serum were detectedas normal contrast.Results①The levels of IL-12 and IFN-γwere significant lower, and the level of LDHsignificant higher(P<0.01) in AL patients before treatment in comparison with the controlgroup. There was a significant positive correlation between the levels of IL-12 and IFN-γ, anda negative correlation between the levels of LDH and IL-12 or IFN-γ(P<0.01). IL-12 inpatients with AML was lower than patients with ALL in preliminary diagnosis, but there wasno significant difference in IFN-γor LDH (P>0.05).②In CR patients the level of serumIL-12 and IFN-γincreased significantly while the level of LDH decreased markedly. Therewere statistic difference in comparison with primary ALL, respectively (P<0.05). In relapsepatients, the level of serum IL-12,IFN-γdecreased and the level of LDH increased, there wasa significant difference in comparison with the CR group (P<0.05).③There were nosignificant differences among different subtypes according to FAB classification in ALs(p>0.05). The level of IL-12 in preliminary B-ALL was higher than that in preliminary T-ALL,and there was significant difference between B-ALL and T-ALL.④The decreased degree ofserum IL-12 and IFN-γin patients who were not sensitive to chemical treatment wassignificant higher than that in patients who were sensitive to chemical treatment, meanwhilethe increased degrees of serum LDH in these two groups were evident.Conclusions①The evidence of decreased serum IL-12,IFN-γ, increased serum LDHand the significant positive correlation between IL-12 and IFN-γin initial diagnosis patientssuggest that there is close correlation between serum IL-12,IFN-γand LDH and themechanism of AL.②The reality of increased serum IL-12,IFN-γand decreased serum LDHin CR patients and the opposite changes of these three factors in relapse period show thatthe levels of serum IL-12,IFN-γand LDH is closely associated with the development of AL.③The decreased degree of serum IL-12 and IFN-γin patients who were not sensitive to chemical treatment was significant higher than that in patients who were sensitive to chemicaltreatment. And the patients with consistant change (increase or decrease) had poorprognosis. These facts show that the levels of serum IL-12,IFN-γand LDH are related withthe treatment of AL and can be regarded as important markers in prognosis.④There is norelation between the levels of serum IL-12,IFN-γand LDH and AL FAB classification. Butthe level of IL-12 in preliminary B-ALL is higher than that in preliminary T-ALL, it showsthat there is relation between the level of serum IL-12 and immunological classification inALL.⑤The change of the level of IL-12, IFN-γand LDH before clinical symptomsappearance in part of CR patients indicates that the detection of IL-12, IFN-γand LDH can beused in early diagnosis in relapse patients and the guidance of clinical treatment. |
