The Study Of Qing Huo Bai Du Yin Affect The Expression Of High Mobility Group Box Chromosomal Protein 1 In Macrophage

Objective: Observe the expression change of HMGB1 in RAW264.7 macrophage after interfered with burning serum and Qing Huo Bai Du Yin serum(QHBDY serum), find out the endogenous protect mechanism of QHBDY resist inflammation reaction .Methods: Interfere the RAW264. 7 macrophage with normal SD rat serum , burning SD rat serum and QHBDY feeding SD rat serum ,use Rui-Shi dyeing to observe the morphological change of cells , MTT test to evaluate the activity of the cells ,and RT-PCR to find out the expression change of HMGB1 in RAW264. 7 macrophage .Results : Use the concentration of 5%, 10% and 20% normal serum, burning serum and herb serum of SD rat to interfere RAW264.7 macrophage ,â‘ different concentration of the same serum can not affect the activity of the RAW264.7(P >0.05) ;â‘¡different serum of the same concentration can reduce the activity of the RAW264.7 following the length of time ,but the herb serum of SD rat affect the activity for the least.There is a significant difference between the three kind of serum(P<0.05);â‘¢the extinction luminosith value of complete medium is higher than mis-classification medium,so the serum of cattle can affect the activity of RAW264.7(P<0.05), it can provide the nutrient for the cell culture.Use the concentration of 20% normal serum of SD rat, burning serum of SD rat and herb serum of SD rat to interfere RAW264.7 macrophage, examine the expression of HMGB1 by reverse-transcription polymerase chain reaction. Discovered thatâ‘ small quantity of HMGB1 expressed in RAW264.7;â‘¡the expression of HMGB1 fluctuate around the standard level after interfered with normal serum of SD rat;â‘¢the expression of HMGB1 rise in 3 hour ,then come down to the standard level. In the 18 hour ,it rose rapidly, at 36 hour it reached the peak, in 48 hour it still remain the high level after interfered with burning serum;â‘£the expression of HMGB1 rise in 3 hour ,then come down to the standard level. In the 24 hour ,it start to rise after interfered with herb serum,it is lower than the burning serum interfere group; Conclusion:â‘ Burning serum can increase the expression of HMGB1 in RAW264.7, it can damage the cell;â‘¡Herb serum of QHBDY can decrease the high expression of HMGB1 in RAW264.7 caused by burning serum, it can protect the cell.
