Objective To study the effect of fatty acids on the expression of adiponectin and adiponectin receptors mRNA in 3T3-L1 adipocytes in vitro.To study the function of adiponectin and its receptors in procession of IR Methods The murine 3T3-L1 preadipocyte cell line(provided by Institute of Biochemistry and Cell Biology,Chinese Academy of Sciences)were grown in DMEM containing 25mmol/L glucose as normal anchoringcells.Confluent preadipocytes were cultured for 2 days in culturemedium further supplemented with 10ug/ml insulin,0.5mmol/L isobutylmethylxanthine and 0.25umol/L dexamethasone and 2 days inculture medium with 10ug/ml insulin.After additional 4 to 6 days inculture medium,more than 90%of the cells had accumulated fatdroplets 3T3-L1 adipocytes were treated with different concentrations range of PA and DHA in vitro.The expression of adiponectin and adiponectin receptors(R1,R2)mRNA were measured by semi-quantitative RT-PCR withβ-actin as internal control.Results Compared with the control group,the expression of adiponectin decreased in the group with PA.When the concentrate of PA was over 200μmol/L,,the expression of adiponectin decreased significantly compared with control group(P<0.05).Expression of adiponectin increased in the group with DHA,but there is no significant difference(P>0.05).R1,R2 expression of 3T3-L1 adipocyte decreased in the group with PA,but there is no significant difference(P>0.05).R1 and R2 expression in the group with DHA have no significant difference compared with control group (P>0.05)..Conclusion Treatment of 3T3-L1 cells with different PA and DHA for 48hours,the effect of DHA on the expression of adiponectin and adiponectin receptors(R1,R2)mRNA is not significant.The effect of PA on the decrease of adiponectin and R1,R2 expression in 3T3-L1 adipocytes may participate in IR induced by FFA.
