Objectives: HCY is an independent risk factor for cardiovascular disease in humans.To investigate the effects of HCY on apoptosis,production of reactive oxygen species(ROS)and expression of caspase3 in HUVEC cultured in high glucose.Methods:HUVECS were cultured in vitro and were divided into four groups:control group;high glucose(HG) group;HCY group;HG with HCY group.The distinction was raising 12h,24h,48h carried on the correlation the experiment and the examination,the activities of SOD and the level of MDA in the supernatant were measured by spectrophotometry.Flow cytometer and agarose electrophoresis of DNA fragmentation were applied to observe cell apotosis and the expression of caspase3mRNA were measured by RT-PCR.Results:Compared with the control group. HCY induced endothelial cell apoptosis in a time-dependent manner and the activities of SOD were markedly lower,while the level of MDA were greatly higher. HCY stimulated the expression of caspase3mRNA (P<0.01) .Also this kind of change was more obvious in the high glucose with HCY group (P<0.01) .Conclusions:HCY might activated apoptoic gene directly or indirectly (partly through nuclear factor-kappa binding approach)through oxidative stress mechanism and up-regulated the expression of caspases,HUVEC happened apoptoic damage,Thus damaged the vascular endothelial cells and aggrevated the diabetes vascular complications.
