Although being some effective, actual anti-carcinoma drugs present a lot of problem such as poor performance and specificity, high toxicity and side-effect, drug-resistance and so on. Chinese medicine has long history, accumulates abundant experience and has idiographic superiority: high performance and specificity and low toxicity, so it becomes a important and promising direction to search anti-carcinoma from Chinese medicine especially toxicity-free Chinese medicine.Toxicity-free Chinese medicines were screened on human chronic myloid leukemia K562 cell, hepatoma HepG2 cell, cervical carcinoma Hela cell and gastric carcinoma BGC823 cell to find anti-carcinoma drugs. Extracts of Cirsium japonicum Fisch.DC, Cirsium setosum (Willd.) MB and Sanguisorba officinalis L were found to be much effective. The extracts depressed growth curve and cloning efficiency of the four cells significantly and made the cells shrinkage, roundness, floating and crumbling which were observed under the inverted microscope. The extracts of Cirsium japonicum Fisch.DC and Cirsium setosum (Willd.) MB could induce apoptosis in HepG2 cell and might relate with down-regulation of mitochondrial membrane potential, which were detected by Hoechst 33342,Annexin-V/PI and rhodamine-123 staining. Extracts of Sanguisorba officinalis L could arrest HepG2 cells at S phase.The different inhibit effects of one extract on four kind cells conformed to the basic theory of Chinese medicine which shows once again that the abundance of basic theories of traditional Chinese medicine can also be exhibited in vitro at cell level. This pattern has important guidance meaning to Chinese traditional medicine research by laboratory methods.A summary including progress in anti-carcinoma Chinese medicine and cell apoptosis was also given in this paper. |
