Objective:To investigate the relationship between the T573C allele polymorphism of angiotensin II type 1 receptor (AT1R) gene and the essential hypertension(EH) in Kazakan of Xinjiang.Combining with the phenotypic data, the possible suspectible marker of EH in Kazakan was determined.Methods:Using randomiazed sampling methods of cross-sectional study, a population-based case-control study was carried out. The population was selected randomly from the pasturing area aged 35 to 65 years in Xinjiang province. Genomic DNA was extracted from the peripheral blood leukocytes by using SK1252 Genome DNA Kit. Polymerase chain reaction and restriction fragment length polymorphism methods (PCR-RFLP) were used to detect the T573C polymorphism of AT1R gene in Kazakan including 197 patients with hypertension and 195 normotensive controls. Through observing the three genotypes distribution of T573C polymorphism between hypertensive group and normotensive group, the possible relationship between T573C polymorphism and hypertension of Kazakan was analyzed.Results:(1)The frequencies of the two alleles,T573 and C573, on T573C polymorphism of AT1 receptor gene and three genotypes of T573C polymorphism are in Hardy-Weinberg eqilibrium.(2)The T573 and C573 allele frequencies were 0.8071,0.1929 respectively in hypertensive group and 0.7256,0.2774 in the normotensive group and T573 allele was more frequent in hypertensive group(χ2=7.269,P=0.007)The odds ratios (OR) was 1.582 and the 95%confidence intervals (95%CI) was(1.132~2.211).(3)The genotypes frequencies of T573C polymorphism were TT 0.6294, TC 0.3553,CC 0.0152 in the hypertensive group and the corresponding frequencies were TT 0.5128,TC 0.4526,CC 0.0615 in the control group respectively, the genotypes distribution were significantly different between two groups (χ2=9.066,P=0.011).TT genotype increased greatly in hypertensive group(χ2=5.442,P=0.02)。(3) CC genotype was not detected in female hypertensives, the distribution of three genotypes composed of T573C polymorphism was different between female hypertensive group and the control group(χ2=9.427,P=0.009).TT genotype was more frequent in hypertensive group(χ2 = 4.80,P = 0.028,OR=1.614,95%CI 1.078~2.415). But no statistical differences was found in male Kazakan(P=0.443).(4) The female Kazakan carrying TT genotype have siginifidantly higher SBP and DBP reading than those who carrying CC genotype(P value were 0.019,0.037 respectively).Conclusion: T573C polymorphism of AT1 receptor gene may relate to the essential hypertension of Kazakan of Xinjiang, and the T573 allele may be one of a genetic suspectibility factors for the hypertension of Kazakan in Xinjiang,but C573C genotype might be a protective fator for the hypertension of Kazakan. Kazakan carrying T573T genotype, especially for female Kazakan are apt to develop essential hypertension.
