| Purpose: The objective of the study is to investigate the acute nephrotoxicity pathologicalchanges of LianBiZhi injection in mice, investigate two drugs combined use whether willincrease the toxic action of the kidney, and inquiry its mechanism of nephrotoxicity.Methods: Select 40 kunming mouse in SPF that the body weight is 20_+ 5g, female and maleeach half, with adaptability raising two days. The mouse were divided into 4 groups atrandom: negative control, positive control, LianBiZhi and combined use groups, each 10 withfemale and male each half. Mouse of each different group were administered discernlyPhysiologic Saline, Gentamicin Sulfate Injection, LianBiZhi Injection, both GentamicinSulfate and LianBiZhi Injection for intraperitoneally injection once a day for 7 days. Allmouse were weighted before administration and the dose was adjusted according to the bodyweight variety. Blood of mouse in each group was taken from carotid vascellum afteradministered intraperitoneally 2%pentobarbital sodium and examined Blood Convent, SerumCreatinine(Cr) and Urea Nitrogen(Urea). The kidneys of mouse were taken to do pathologicalexamination after sacrificed of the animals.Results: 1. The main acute nephrotoxicity pathological changes of LianBiZhi injection arerenal tubular epithelial celluar swelling, and basophilia degeneration in the corticomedullaryjunction. These are different from the renal tubular epithelial cells analosis in the superficialpart of the renal cortex and renal tubular epithelial cells vacuole apomorphosis, necrosis in thecorticomedullary junction of the positive control group; The pathological changes of thecombined use group are same alike to the positive control group but the degree is moreserious; 2. The positive control and combined use groups compared with the negative control group, the expression of VEGF, VEGF-R1, VEGF-R2 elevated equally, with significantdifference (P<0.01); The expression ofVEGF, VEGF-R1, VEGF-R2 in LianBiZhi grouphad not marked changes, without significant difference (P>0.05).Conclusions: 1. Short-term large quantity use LianBiZhi injection will induce acutenephrotoxic action in Kunming mice, the toxicological target organ is the renal tubules in thecorticomedullary junction, comparing with aminoglycoside antibiotics, the degree ofnephrotoxicity is lower; 2. Two drugs combined used have congenrous nephrotoxic action,the toxicological target organ is the renal tubules in the whole renal cortex, and the degree ofinjury in the corticomedullary junction is more serious; 3. The nephrotoxic mechanism ofLianBiZhi injection is main the directly toxic action of renal tubular epithelial cells;when twodrugs combined use, because of the two drugs all having the direct nephrotoxic action thatcaused the toxic effect added, So the nephrotoxicity is more aggravation than single druguse. 4. the expression of VEGF, VEGF-R1, VEGF-R2 elevated in the positive control andcombined use groups that can cue the mechanism of renal injure relate to hypoxia probably. |
