Experimental Study Of The Cholesterol Calculus Dissolved By Methyl Tertiary Butyl Ether(MTBE) Together With Bletilla | | Posted on:2008-04-08 | Degree:Master | Type:Thesis | | Country:China | Candidate:M J Lu | Full Text:PDF | | GTID:2144360215965338 | Subject:Integrative Medicine Surgery | | Abstract/Summary: | PDF Full Text Request | | Objective:Insert gallstone to rabbit gallbladder to establish the model of cholelithiasis by experiment, survey the protective efficacy and the mechanism of Bletilla to the damage of gallbladder, hepatic function and renal function induced by MTBE.Methods:rabbits 24 were randomly allocated to four groups[ NS group, MTBE control group, experimental group (high dosage Bletilla and MTBE group) , experimental 2 group(low dosage Bletilla and MTBE group) ]. Every group has six rabbits. Inject 3% sodium pentobarbital(1ml/kg) to anaesthesia rabbits and retained the rabbits on immobilizers. Segregate the trach and tracheal intubation to prevent rabbits' death. Split and open the chest to exposure gallbladder fully. Shear a microstomia on collum vesicae felleae and then insert a gallstone in the gallbladder after the bile eliminate literally. Insert a vessel and suture the microstomia. Another vessel was insert in bottom of blastula on the same way. Gallbladders were flush with NS after no leakage was assured and the NS outflowed from the other vessel. Gallbladders of NS control group were infused NS(2ml) every thirty minutes. This perfusion lasted for ten hours. MTBE group gallbladders were infused MTBE (2ml) every thirty minutes and lasted for ten hours. High dosage Bletilla and MTBE group gallbladders were infused high dosage of Bletilla(1g/2Oml, 2ml) every thirty minutes and lasted for three hours. After that, MTBE was infused in the gallbladders (2ml) every thirty minutes and lasted for ten hours. The same treatment were done on the low dosage Bletilla and MTBE group gallbladders as high dosages of Bletilla group. Each time after infused liquid to gallbladders the two vessels were clamp by bulldog clamps to make sure that the gallbladders were engorged. Breathing frequency and heart rate were surveyed during the experiment. After ten hours of perfusion the remnant stones were took out and weighed after dried. And then calculated the ratio of gallstone dissolution. The gallbladders and hepar tissue nearby the gallbladders were fixation witn 10% formaldehyde. Blood was draw-off from carotid artery before and after the experiment, segregation and got blood serum to measure amount of uric acid(UA), urea nitrogen(UN), creatinine(Cr), and activity of ALT, AST, GGT, AST/ALT. gallbladders and hepar tissue nearby the gallbladders were overviewed under light microscope.Results:Ratio of gallstone dissolution of MTBE group, high dosage Bletilla and MTBE group , low dosage Bletilla and MTBE group were obvious higher than NS group(P<0.01). And the differences of MTBE group, high dosage Bletilla and MTBE group, low dosage Bletilla and MTBE group had no statistical significance(P>0.05). Compared with NS group, the scale of other three groups blood serum UA, UN, Cr had no statistical significance(P>0.05). After experiment, AST, GGT activity rose a little but had no statistical significance vs NS group (P>0.05). MTBE group blood serum ALT activity was distinguished higher than NS group(P<0.01), high dosage of Bletilla could markedly depress the rice of ALT activity induced by MTBE(P<0.05). After experiment, each group blood serum AST/ALT had depressed a little but the depression had no statistical significance vs NS group(P>0.05).Reviewed under light microscope NS group mucous membrane of gallbladder were integrity, no damage was seen. (fig. 8). MTBE group mucous membrane of gallbladder had massive abscission and inflammatory cell(Fig. 9, Fig. 10). Only a little neutrophilic granulocyte can be seen in discrete mucous membrane of gallbladder in high dosage Bletilla and MTBE group(Fig, 11, Fig. 12). Some exuviations and neutrophilic granulocyte could be seen in low dosage Bletilla and MTBE group(Fig. 13, Fig. 14). Hepar tissue nearby the gallbladders of MTBE group had some inflammatory cell infiltration(Fig. 16). Just a little inflammatory cell infiltration can be seen in hepar tissue of low dosage Bletilla and MTBE group(Fig. 18).
