After a long period of clinical practice of our people, it has become the historical legacy. With the development of science and technology, the way to explore modernization research of Chinese traditional medicine, and establish standardized fingerprint database, has become an important task in Chinese traditional medicine research. Fluorescence analysis has become an important method in the study of Chinese traditional medicine and its active components. In particular, the development of three-dimensional fluorescence technique provides a new way to the fingerprint identification of Chinese traditional medicine.In this thesis, fluorescence spectra of water extraction of three certified pharmacopeia rhubarbs have been studied. An operation for extraction and separation of anthraquinone compounds from a rhubarb sample has been done, and a preliminary analysis of three-dimensional fluorescence spectra of rhubarb has been given.The main contents of the thesis are as follows:1. Three certified rhubarb samples were studied by fluorescence method in different experiment conditions. It was found that Rheum palmatum L. and Rheum officinale Bail had very similar fluorescence spectra, but Rheum tanguticum Maxim. Ex Balf was obviously different. The spectral feature can be used in the qualitative identification of them. As for the quantitative determination of the fluorescence components, good linear relationships between fluorescence intensity and concentration have been established.2. Anthraquinone compounds were studied by fluorescence method in different experiment conditions. It was discovered that the characteristic fluorescence peaks of free anthraquinone compounds were located in the vicinity ofλex/λem = 450nm/540nm. According to their spectroscopic properties change with acidity, the ionization constants of rhein,emodin and aloe emodin were determined by UV and fluorescence methods respectively. Using quinine bisulphate as a reference, fluorescence quantum yield of aloin was measured to be 0.053.3.
