Research Of Vitamin E Succinate Induce Apoptosis In Prostate Cancer PC-3 Cells

OBJECTIVE:To study the apoptosis of PC-3 cells induced by Vitamin E succinate(VES) and its possible molecular mechanisms. MRTHODS:The PC-3 cells were cultured in vitro within VES of different concentration.The proliferation of PC-3 cells were determined with an MTT colorimctric assay was used to examine morphological changes of apoptosis.The cell cycle and expression of Fas were analyzed by fiow cytometry(FCM).The expression of TGF-βwas measured by an ELISA assay. RESULTS:The proliferation of PC-3 cells were inhibited significantly by VES. FCM indicated the increased cell population in the G2/M. phase was associated with a decreased cell number in the S phase.Expression of Fas protein and TGF-βincreased significantly.Conclusion:VES can induces apoptosis in prostate cancer PC-3 cell . Fas protein and TGF-βmay play important roles in the molecular mechanisms.
