Different techniques for the determination of uric acid and creatinine have been developed and applied in clinic laboratories.In this thesis, an ID/LC/MS method was developed to determine uric acid and creatinine in serum. [1, 3-15N2]uric acid and D3-creatinine were selected as labeled materials in the research. The conditions in serum sample pretreatment and the operation parameters of LC/MS were studied.At the optimized condition, the RSD of the uric acid and creatinine of three human serum samples by the ID/MS are less than 1%. Two NIST standard samples SRM 909b (â… ,â…¡) were used as certificated standards to evaluate the established method. The result showed that relative diviationes were less than+1%. The results obtained by this method were compared with that obtained by different columns and by LC/MS/MS with relative deviation less than±1%. The recoveries ranged from 99% to 101%. The total uncertainty of uric acid and creatinine of four human serum samples assays are less than 1%. The results showed that it could be hopefully recommended as reference method for uric acid and creatinine assays.
