Objective: To investigate astragalus membranaceus on the immune disorder (Th1/Th2 Disequilibrium) in rat with IgA nephropathy.Methods: Rat IgA nephropathy (IgAN) model was duplicated by oral feeding of bovine serum albumin (BSA) and carbon tetrachloride (CCL4) hypodermic injection and lipopolysaccharide (LSP)vena caudalis injection. The rats were divided into three groups randomly for the normal, IgAN model group and the group treated with astragalus membranaceus. The treatment group was given the astragalus membranaceus medicines for intragastric administration, the normal group and the IgAN model group had been gave the coequal perfusion of aqua destillata. The rats was examined with albuminuria , haematuria and pathological change of renal tissue and examined the distribution of TGF-beta and interleukin- 5 in renal tissue by immunohistochemistry technology and the INF-γand IL-4 of cytokine of Th1 and Th2 was detected in rat IgA nephropathy model by Sandwich enzyme linked immunosorbent assay (ELISA).Results: (1) The establishment model to succeed . There are the fairly clumping flavovirens IgA fluorescence or the granular IgA fluorescence medium intensity in the region of Mesangium of IgAN model group, means the duplication of model rat to succeed. (2) The influence of astragalus mongholicus on cruenturesis and urine protein: The urine red cell count (URCC) were 599.9±4.16 piece /ul,16.91±1.16piece/ul,14.33±1.61 piece /ul of the IgAN model group, the astragalus mongholicus group and the normal group, obiviously the compared URCC of urine of model IgAN were higher than the astragalus mongholicus gropu and the normal group. The albuminuria were divided 55.91±0.99mg/L,14.16±0.56mg/L,13.37±0.89mg/L of model IgAN, astragalus mongholicus and normal group. The 24 hours ruine protein of model IgAN were obviously higher than the astragalus mongholicus group and the normal group.(3) The influence of astragalus mongholicus on nephridial tissue pathology of experimental rat: The injury of glomerular mesangium; nephric tubule and renal interstitium pathology were compared gravely than the astragalus mongholicus group and normal group, there were comparely strong dumping flavovirens IgA fluorescence or granular medium intensity of IgA fluorescence in the glomerular mesangium of rat of model IgA.The IgA fluorescence was bated obviously in the glomerular mesangium of astragalus mongholicus cure's group. The expression were reflected in the sporadic line-form and granular IgA deposit lightly in glomerular mesangium, the glomerular mesangium of model group no IgA deposit Kind of some statistical meaning existed in the IgA immune fluorescence intensity of glomerular mesangium of model IgAN and the astragalus mongholicus cure's group.(4) The influence of astragalus mongholicus on TGF-β1 and IL-5 of nephridial tissue of experimental rat: The expression surface were 62.30±1.62%,27.51±8.62%,26.81±8.13%, the expression of TGF-β1 of renal tissue of IgAN model were obviously strengthend, his meaning were more valuable in statistical than the astragalus mongholicus cure's group (P<0.01) and normal group (P>0.05) , expression surface were 64.39±2.97%,28.41±5.20%,26.74±1.37% of model IgAN, Astragalus mongholicus cure's group and normal group, nephridial tissue IL-5 of model IgAN were obviously strengthened, his meaning was more valuable than the Astragalus mongholicus cure's group (P<0.05) and the normal group (P<0.05) , there is no statistical meaning between the Astragalus mongholicus cure's group and normal group.(5) The influence of Astragalus mongholicus on IL-4 and INF-γof serum of experimental rat: The level of IL-4 of model IgAN, astragalus mongholicus cure's group and normal group were 35.29±0.84 pg/ml,3.19±1.08 pg/ml,2.75±0.36pg/ml pg/ml,the serum IL-4 of model IgAN was ascended obviously, his statistical meaning were more valuable than the astragalus mongholicus cure's group (P<0.05) and normal group(P<0.05) , there is no statistical meaning between the astragalus mongholicus cure's group and normal group(P>0.05). The level of INF-γwere 16.19±0.66 pg/ml,39.79±1.00 pg/ml,40.30±0.80 pg/ml of model IgAN, astragalus mongholicus cure's group and normal group, the level of serum of model IgAN were descended obviously, his statistical meaning were more valuable than the astragalus mongholicus cure's group (P<0.05) and normal group (P<0.05) , there is no statistical meaning between the Astragalus mongholicus cure's group and normal group (P>0.05) .Conclusions: The astragalus mongholicus can lower the level of hematuria and 24 hours-albuminuria of model IgAN, and ameliorate the change of the renal pathology and bate the deposit of glomerular mesangium. The possible mechanism is to regulate the derangement of Th1,Th2, accordingly to improve the level of IL-4 and INF-γof the cytokine of serum and diminish the expression of cytokine Th2 TGF-β1 and IL-5 of the nephridial tissue, to postpone the development of IgAN.
