| Objective: Investigated the expression of CD59 and DAF mRNA inperipheral blood of MG patients, analyzed the correlation betweenanti-AChR-IgG and this two complement regulator. To study the role ofCD59 and DAF in the pathogenesis of MG and hope finding a newteatment of MC.Methods: 41 patients with MG and 20 healthy controls (HC) wereinvestigate by retrospective study. The expression of CD59, DAF mRNAin peripheral blood mononuclear cells (PBMNCs) were detected byRT-PCR and the levels of serum anti-AChR-IgG were detected by ELISA,the relationship between the anti-AChR-IgG and CD59/DAF wereanalyzed.Results: (1) Expression of CD59 and DAF mRNA in PBMNCs was0.20±0.05 and 0.25±0.07 in MG group, 0.41±0.09 and 0.44±0.12 in HCgroup, respectively. CD59 and DAF m RNA levels in PBMNCs weresignificantly decreased in MG patients compared with HC (P<0.01). (2)Expression of CD59 and DAF mRNA in PBMNCs was 0.20±0.03 and0.25±0.04 in 25 patients with ocular MG (oMG), 0.21±0.03 and0.25±0.05 in 16 patients with generalized MG (gMG),respectively. Therewas no significant difference between the oMG and gMG (P>0.05). (3)The OD value of serum anti-AChR-IgG in MG group was 0.53±0.17.There was no correlation between expression of CD59/DAF mRNA in PBMNCs and and-AChR-IgG level (P=0.961, 0.894).Conclusion: (1)Decreased expression of CD59 and DAF mRNAmay play a critical role in the pathogenesis of MG. (2)There are nodifference in CD59/DAF mRNA expression between oMG and gMG.(3)There was no correlation between the levels of serum anti-AChR-IgGand the CD59/DAF in MG patients.They may participate in differenthumoral immunity passageway. |
