Objectives:1. To investigate the effect of PMA on COX-2 expression in HaCaT cell line, analyze the role of COX-2 in the mechanism of PMA -induced tumorigenesis.2. To investigate the effect of TP on COX-2 expression in PMA-treated HaCaT cell line and A431 cell line, analyze the mechanism of treating SCC by TP.Methods:1. The expression of COX-2 mRNA and protein in HaCaT cell line treated with different concentrations of PMA was studied by reverse-transcription polymerase chain reaction and Western-blot. We compared COX-2 mRNA and protein expressions between different teams.2. The expressions of COX-2 mRNA in TP-treated A431 cell line and TP+PMA-treated HaCaT cell line were studied by reverse-transcription polymerase chain reaction.We compared COX-2 mRNA expressions between different teams of A431 cell line.We compared COX-2 mRNA expressions between TP+PMA-treated and PMA-treated HaCaT cell line.Results:1. HaCaT cell line didn't express COX-2 mRNA or protein. PMA could induce HaCaT cells to express COX-2 mRNA and protein with a dose-dependent manner under 24 hours co-culture, and there were significant differences by stastics analysis(p<0.01).2. TP could inhibit the COX-2 mRNA expression in PMA-treated HaCaT cell line under 24 hours co-cultue, and there was significant difference by stastics analysis(p<0.01).3. A431 cell line overexpressed COX-2 mRNA. TP could inhibit the COX-2 mRNA expression in A431 cells with a dose-dependent manner under 24 hours co-culture, and there were significant differences by stastics analysis(p<0.01). Conclusions:1. HaCaT cell line didn't express COX-2 mRNA or protein. PMA could induce COX-2 mRNA and protein expression in HaCaT cell line, which suggested that overexpressed COX-2 was correlated with PMA -induced tumorigenesis.2. A431 cell line overexpressed COX-2 mRNA, which suggested that overexpressed COX-2 was correlated with tumorigenesis of SCC.3. TP could inhibit COX-2 expression in A431 cell line and PMA-treated HaCaT cell line, which suggested that TP could treat squamous cell carcinoma by inhibiting COX-2 expression.
