Objective: To implanting electrode in hippocamp of Penicillin-induced rats, observe the changes of the behavior,the electroencephalogram(EEG) of rats and the expression of c-fos,c-jun in rat't hippocamp after transferring electric discharge by electrode, To study the effects and the mechanism of transferring electric discharge on Penicillin-induced seizures.1. Methods:Acute chemical seizure models were made by using microinjection in right hippocampus of rats with penicillin. 36 SD rats were divided into 6 groups randomly with each one having 6 rats and named as following:Group A: Only be induced to seizure chemically, no electrode implantation.Group B: After being induced to seizure chemically, epileptic discharge be transferred out of brain by connecting one end of electrodes implanted in brain with themselves muscles of skull outside brain by wires.Group C: After being induced to seizure chemically, epileptic discharge was transferred from hippocampus in the right side to hippocampus in the left side by connecting two ends of electrodes implanted in brain.Group D1: After being induced to seizure chemically, one end of electrodes used as leading electrodes were implanted in epileptic foci of hippocampus, and the other one was connected with the recepting electrodes of group D2.Group D2: After being induced to seizure, one end of electrodes used as recepting electrodes were implanted in epileptic foci of hippocampus, and the other one was connected with the leading electrode of group D1.Group F: The control group with electrodes implanting in the right hippocampus and not being induced to seizure chemically.All groups of rats' behavior changes were evaluated at 2h, 3h, 4h according to Racine grade, and EEG was recorded at 2h after seizure having been induced. The rats were Killed at 4h, and brains were taken out to test expression of Cxs(Cx26, Cx32, Cx43)by making frozen slide with immunohistochemistry, and then statistical comparations were carried out.Results:1. Comparisons in behavoirs:(1)The tonic-clonic seizure was the major presentations according to Racine grade expect for group F.(2) The frequency of epileptic burst decreased with time's lasting. after seizure having been indued, but there was no significant difference between group A and B, C, D1 and D2.(3)The epileptic seizure bursted after electrodes were connected with wire between group D1 and D2 5-10 minutes later, but was little slighter in amplitude in group D2 than D1. While in group F, no seizure burst could be found.2.Comparison in EEG:(1)There was no epileptic EEG change in group F.(2) The frequency of epileptic EEG burst in group B was less than group A in 2h (P<0.05).(3)There was no significant difference in frequency of epileptic EEG burst between group D1 and D2(P>0.05).3.The expression of c-fos,c-jun:(1) The expression of c-fos,c-jun in right hippocamp of rats implanted electrode was more lower than the group A, P<0.05, which hinted that the electrode could transfer the epileptic discharge of rats with seizure.(2) The expression of c-fos,c-jun in amphi-hippocamp of group A had no difference, which hinted that the electrode could transfer the epileptic discharge of rats to the other side and make the expression of c-fos,c-jun increased.(3)The expression of c-fos,c-jun in right hippocamp of rats between group D1 and group D2 had not difference, P>0.05, and the expression of c-fos,c-jun in right hippocamp of group D2 was more higher than the group F, P<0.05, which hinted that epileptic discharge of rats could be transferred to left or other rat's hippocamp and make the expression of c-fos,c-jun increased.(4) The expression of c-fos,c-jun in right hippocamp of group ABD1D2 was higer than the left side. which hinted that The expression of c-fos,c-jun in hippocamp was different between two side in penicillin-induced seizure.Conclusion: 1. Expression of Cx26 and Cx 32 could be down-regulated with the implantation of electrodes in epileptic foci, and it suggested that epileptic discharge could be conducted along electrode and its spread could be weakened in brain tissue.2.The epileptic discharge could be transferred out of the origined brain, and could be conducted to another biological tissue purposefully. It suggested that transfer of epileptic discharge could become a new skill to antiepileptic therapy.
