Objective:To detect the expression of HIF-1α,Survivin,Ki-67 and HIF-1αmRNA in prostatic proliferation and prostatic carcinoma and investigate their roles and mutual correlation among these markers in pathogenesis,progression of prostatic carcinoma,and provide with reference for clinicopathological diagnosis and evaluation of prognosis.Methods:The expression of HIF-1α,Survivin and Ki-67 in proliferative lesion of prostate (15 cases)and prostatic carcinoma(62 cases)was detected by immunohistochemistry method. The expression of HIF-1αmRNA was examined by in situ hybridization.We assessed the relationship among the expression of HIF-1α,Survivin and Ki-67 and clinicopathological characteristics in prostatic carcinoma and assessed mutual correlation amomg the three markers.Results:1.HIF-1αwas expressed in the prostatic carcinoma(69.4%).The plasma of tumor cells showed brown colour by immunostaining but the prostatic proliferation was negtive(0%)(P<0.001).HIF-1αmRNA expression was primarily localized in the plasma of tumor cells. HIF-1αmRNA was positive in the prostatic carcinoma(75.8%),prostatic proliferation was negtive(0%)(P<0.001).2.The expression of HIF-1αand HIF-1αmRNA in prostate carcinoma was significantly related with the histological grading,clinical staging(P<0.05).3.Survivin protein expression was localized in the plasma.Survivin was more expressed in the prostatic carcinoma(71.0%)than in the prostatic proliferation(6.7%)(P<0.001).The expression of Survivin in cancer tissues was significantly related with the histological grading, clinical staging(P<0.05).The expression of Survivin is stronger in poorly-differentiated prostatic carcinomas(100.0%)than that in well-differentiated(57.1%),moderate-differentiated prostatic carcinomas(64.0%)(P<0.05).But there was no significant difference between Survivin expression of well-differentiated and moderate-differentiated prostatic carcinomas(P>0.05).4.Ki-67 expression was localized in the nucleus.Ki-67 was expressed in the prostatic carcinoma(64.5%)but not in the prostatic proliferation(0%)(P<0.05).The expression of Ki-67 in cancer tissues was significantly related with the histological grading,clinical staging(P<0.05). 5.The expression of HIF-1αwas correlated with expression of Survivin and Ki-67.The value of r_s was 0.377,0.362 and 0.391 respectively.Conclusions:1.The high expression of HIF-1αmRNA and HIF-1αprotein in prostatic carcinoma indicates that HIF-1αmight be a key gene,which induces the occurrence and development of prostatic carcinoma.Detecting the level of HIF-1αexpression can be an important measure for monitoring the defter and prognosis of prostatic carcinoma.2.The high expression of Survivin and Ki-67 in cancer tissues was significantly related with the histological grading,clinical staging.It suggests that the occurrence and development of prostatic carcinoma were related with the cell apoptosis and proliferation.3.There was a positive correlation among HIF-1α,Survivin and Ki-67 in prostatic carcinoma,which showed that HIF-1αmight play an important role during the occurrence and development of prostatic carcinoma by stimulating the proliferation and apoptosis of tumor.4.It may be a new way to treat prostatic carcinoma by inhibiting the transcription activity and expression of HIF-1α.
