Objective:To measure the proportion of monocyte-macrophage(CD64~+)in AA patient's bone marrow,the expression of RCAS1 mRNA in bong marrow mononuclear cells(BMMNC),and the influence of RCAS1 to colony forming unit-erythrocyte(CFU-E),colony forming unit-common precursor of granulocyte and macrophage(CFU-GM);evaluate the monocyte-macrophage(CD64+),RCAS1 and their influence to the hematogenesis;explore the role of MPS in the pathogenesis of AA.Methods:①The quantity of monocyte-macrophage(CD64~+)in bone marrow of AA patients in diagnosis and normal controls were detected by FACS;②mRNA expression of RCAS1 in BMMNC from AA patients and normal controls were measured by reversetranscriptase polymerase chain reaction(RT-PCR),③Bone marrow stromal cells were cultured,the macrophage were enhanced and stimulated;④The affect of RCAS1 in colony forming uniterythrocyte (CFU-E),colony forming unit-common precursor of granulocyte and macrophage (CFU-GM)were measured.Results:①In aplastic anemia patients'BMMNC,the percentage of CD64+ cells were (13.10±8.07)%.In normal controls,they were(8.54±4.40)%,respectively.In the analysis of bone marrow morphology,the proportion of monocyte was(1.42±2.80)%in patients and (0.67±0.82)%in normal controls.The percentage of CD64+ cells and the proportion of monocyte were significantly higher than those of controls.②The positive rates of RCAS1 of AA patients was 12/22.In normal controls,it was 5/25.And the expression intensity of RCAS1 was higher in AA patients than in normal controls.③Enhanced and stimulated by M-CSF,LPS, the macrophage which is from bone marrow stromal cell culture and the expression of RCAS1 mRNA were higher than controls.④The number of CFU-E were reduced by RCAS1,(the experiment group with sRCASlwere 33.0±7.84,while the controls were 63.2±6.22, P<0.05);but no influence in CFU-GM,(the experiment group with sRCAS1 were 43.4±5.94, while the controls were 48.0±3.54,P>0.05).And these phenomena were not depended on Fas-FasL,(the group with Fas-Ab were 33.0±7.84,and the controls without Fas-Ab were 32.8±5.12,P>0.05).⑤The expression of RCAS1 were negatively correlated with their RC.Conclusions:The proportion of monocyte-macrophage(CD64~+)in AA patient's bone marrow increased,predominant expression of RCAS1 in stimulated macrophage might lead to the apoptosis of erythroid progenitor cells.
