Objective:In this study the model of nephritic syndrome (NS) was made with puromycin aminonucleoside (PAN). The NS rats were cured by Shen-Le capsula mollis. Proteinuria plasma protein, blood-fat, renal function, renal pathomorphology, the expression of renal TGF-β1 were observed. The effect and mechanism of Shen-Le capsula mollis on NS were investigated so as to provid experimental data.Method:Sixty male Wistar rats were selected to our experiment. After one week, they were put into metabolism cage to obtain urine, and urine protein and red blood cells were detected. The result was negative. Then they were randomly divided into six groups: Normal, Model, TWP group and low-dosage shen-le group, middle-dosage shen-le group, high-dosage shen-le group (n=10, in each group). Each animal adopted tail intravenous injection of PAN according to 50mg.kg-1 except the Normal group. PAN was injected according to 5mg.kg-1 at the 13,16,19 day of injecting the first PAN. Drugs were administered from seventh day after duplicating model but the saline were used in Normal and Model groups orally. 24h urine protein was determined with the trichloroacetic acid turbidimetry. All animals were killed at the 31th day. SerumTP, Alb, TC, TG, BUN, Scr were assayed with the automaticbiochemistry analyzer. Blood viscosity, hematoplasma viscosity and volume packed cells were detected by blood viscosity instrument. Each group of glomerular basement membrane and foot process were observed with light microscope, electron microscope. The expression of TGFβ1 was observed with the immunohistochemical staining. The sectionof HE, Masson, PAS staining and immunohisto- chemistry staining was analyzed with the pathology image analysis system.Result: (1) 24h urinary protein quantitative analysis showed that: At the fifth, tenth, fifteenth, twentieth, twenty-fifth, thirtieth, the Model group was significantly higher than Normal group (P<0.01). Model group urine protein compared with four treatment groups had the unusual significant difference except for low-dosage shen-le group (P<0.01), though the low-dosage shen-le group urine protein is lower than the model group, but there was no significant difference between them (P>0.05). high-dosage shen-le group urine protein compared with TWP group had no statistics significance (P>0.05).(2) At the 31th day each group of biochemistry analysis showed that: TP and Alb of the blood serm in Model group was descent significantly than Normal group (P<0.01); TP and Alb in other three treatment groups increased significantly than Model group except for low-dosage shen-le group (P<0.05), though the low-dosage shen-le group urine protein is lower than the model group, but there was no significant difference between them (P>0.05). There was no significant difference between high-dosage shen-le groups and TWP group (P>0.05). TC and TG of the blood serm in Model group increased significantly (P<0.01), TC and TG of the other treatment group was decent than Model group except for low-dosage shen-le group (P<0.01), though the low-dosage shen-le group TC and TG is lower than the model group, but there was no significant difference between them (P>0.05). There was no significant difference between high-dosage shen-le groups and TWP group (P>0.05). BUN and Scr of the blood serm in Model group were higher than Normal group significantly (P<0.01); BUN and Scr of other treatment groups decented significantly than Model group except for low-dosage shen-le group (P<0.01), though the low-dosage shen-le group BUN and Scr is lower than the model group, but there was no significant difference between them (P>0.05). There was no significant difference between high-dosage shen-le groups and TWP group (P>0.05).(4) Nephridial tissue pathomorphology showed that: Renal tissue pathomorphology: In the model group, the light microscope observation result showed that: A few of glomerulus manifested segmental glomerulosclerosis, proximal epithelia cell degenerated and swelling; there were proteinic cast and broadened in renal tubular and scatter fibroplasias in interstitial; the electron microscope observation showed that: foot process confluence extensively in glomerulus epithelial cells. The low-dosage shen-le group is homoioplastic with the model group. Though in the high-dosage shen-le group, the changes were better than those in the model group. In TWP group, renal tissue pathomorphology was similar to the high-dosage shen-le group.(5) The expression distribution and semi-quantitative analysis of TGF-β1: In the model group, the relative amount and density of TGFβ1 in glomerulus and renal tubule had significant difference with the compared of the Normal group TGF-β1 had strong expression (P<0.01). But the expression of TGF-β1 diminution in the three treatment groups was lower than model group (P<0.01), except for the low-dosage Shen-le group. And there were no significant difference between the high-dosage Shen-le group and the TWP group (P>0.05).Conclusion: 1 Shen-Le capsula mollis can reduce rat's proteinuria, increase plasma protein, improve blood rheology, decrease BUN and Scr to protect renal function. 2 Shen-Le capsula mollis can improve the clearance ability of the oxygen-derived free radidicals, inhibitor lipid peroxidation, diminution the expression of TGF-β1 to postpone the process of renal pathomorphology. 3 Shen-Le capsula mollis may be similar to TWP in the effect of treating NS.
