Rats were injected 10% chloraldurat in abdominal cavity to anesthetize and cut didymus off in all groups except control group. After one week except control group all groups were injected androlin 0.5mg/0.1ml in each rat, the control group were injected isotonic Na chloride 0.1ml each once daily. After 10 days 12 rats were take from control grouop and model group. They were executed and removaled prostata to calculated prostate index.The control group rats'prostate index is 0.14±0.02,and the model group is 0.22±0.02 (P<0.01)that indicated the model of rat Benign Prostate Hyperplasia was successful.Except control group all groups were injected androlin 0.5mg/0.1ml in each rat, the control group were injected isotonic Na chloride 0.1ml each once daily in 30 days.while Proscar group rat were drenched Proscar 0.45mg/kg, and QLAT group were drenched QLAT as 15 times as adult dose, while controlgroup and model group were drenched distilled water each day. After the last administration 12 hours all of the rats were executed.Each rat'prostata were dissected and surveyed prostate volume and calculated prostate index; using HE dyeing, Prostata were observed in light microscope;using immunohistochemistry we observed the expreesion of PCNA Bcl-2andBax.Results:1 The influence about prostate humid weight prostate volume and prostate index in each groupRat weight has no significant deviation (P>0.05)that show that each grop has comparability. Compared with model group, control group Proscar group and QLAT group's prostate index are obviously low(P<0.01);In Proscar group prostate index (0.77±0.57) has no difference with control group(P>0.05);In QLAT group prostate humid weight prostate volume and prostate index (0.85±0.31)has no difference with control group and Proscar group(P>0.05).2 The influence about rat parastata in pathologyControl group: Prostate glandular organ line up in order, cellula epithelialis are simple columnar epithelium, cell nucleus arrange unilayerly along the bottom of the cell, part cellula epithelialis are more than one unilayer, internal surface of glandular cavity is smooth and glossy. There is no excreta in glandular cavity, part of glandular epithelium make corpora mammillaria enter in the glandular cavity. There is few vessel dilatation.Model group: Prostate glandular organs are tight arrangement, cellula epithelialis are high pillar. Cellula epithelialis are more than one unilayer,most of cellula epithelialis form convex papilate.There is excreta in glandular cavity.There is blood vessel dilatation.QLAT group: Convex papilate are less than model group , lumen of gland become big, cellula epithelialis are unilayer pillar, ,there is no excreta in glandular cavity. There is some blood vessel dilatation.Proscar group: Lumen of gland become smooth and big,the number of cellula epithelialis decrease,cellula epithelialis are simple columnar epithelium. There is few blood vessel dilatation.3 The influence about the expression of PCNA Bcl-2 and BaxThe expreesion of Bcl-2 PCNA is highst in model group ,compared with control group ,there is significant difference(P<0.01).In control group the expreesion of Bax is highst, compared with model group it has significant difference(P<0.01).In QLAT group Bcl-2 PCNA are lower than model group,while Bax is higher (P<0.01); In QLAT group PCNA is higher than control group(P<0.05) and Bcl-2,Bax are no significant difference(P>0.05); In Proscar group PCNA are lower than QLAT group(P<0.05) and Bcl-2,Bax are no significant difference(P>0.05),and the expression of PCNA Bcl-2 and Bax are no significant difference with control group(P >0.05).Conclusions:1 In model group prostate humid weight prostate volume and prostate index is significant higher than control group that show the model of rat Benign Prostate Hyperplasia is successful.2 QLAT can cut down prostate humid weight prostate volum and parastate index number to treat BPH, and the curative effect has no significant difference with Proscar.3 The treatment of QLAT may degrade the PCNA and Bcl-2 while sublime the expreesion of Bax to step down generation activity of cells and promote apoptosis. | | Keywords/Search Tags: | BPH, prostate index, Apoptosis related gene:PCNA,Bcl-2,Bax,apoptosis, QLAT | PDF Full Text Request | Related items |
