Ulcerative colitis, also named nonspecial ulcerative colitis, is a kind of chronicinflammatory disease occurred in recta and cola without the certain pathology.'kuijiefang' decoction is one kind of traditional Chinese medicine formulae that cureulcerative colitis in clinic for many years.Objective: To observe the effect of 'kuijiefang' decoction on rats ulcerative colitis modelswhich is established by 'the compound method of immune induction and chemicalstimulation'; To analyze the functions and the effects of the abnormality of oxygenmetabolism and the level of NO, iNOS in the pathogenesis of ulcerative colitis; To studythe possible target and mechanism of 'kuijiefang' decoction in curing ulcerative colitis.Methods: Wister rats were divided randomly into 6 groups, Group B, M, C, L and I. H..Ulcerative colitis model was established in all groups except Group B by using 'thecompound method of immune induction in combination with chemical stimulation'.General conditions and pathological changes were observed. After 'the model wasestablished, Group B and M were administered normal saline intragastrically. Group C, L,I and H were administered 1.2% SASP, 'kuijiefang' decoction of different concentrations(12%, 68%, 124%) respectively, 5ml/day.per rats, for 4 days. The samples of colon tissueand serum were taken at the end of the test to observe the pathological changes in colontissue, MDA, SOD, NO, iNOS level in serum and colon tissue, and compare and analyzethe differences between the groups. In the method of Pearson's correlation test, analyzedcorrelation between MDA, SOD, NO, iNOS level in colon tissue and serum and score ofgrade of activity of ulcerative colitis.Results: Pathological change in colon tissue of rats of Group M is close to change ofulcerative colitis in human. Rats presented severe diarrhea, even bloody stools, mucousstools, pus stools at the same time. Their general conditions showed a declining trend,their weight was decreased either. Comparing with Group B, Group M's NO, iNOS levelin colon tissue increased significantly (p<0.05, p<0.01). However, there is no statisticallysignificant difference in serum level of NO between Group M and Group B. Comparingwith Group B, MDA, SOD level in the colon tissue and serum increased significantly in Group M (p<0.05). After administration, the indications mentioned above returned totheir normal conditions in Group C, I, H respectively. There was no statistical differencebetween them and Group B. The pathological changes of colon tissue in Group L failed tobe improved significantly, and their general conditions can not achieve the level of GroupB. The abnormal changes mentioned above were remained. There was no statisticaldifference between Group L and Group M(p>0.05). The analysis of correlation betweenscore of grade of activity of ulcerative colitis and serum, level of MDA, SOD, NO, iNOSin colon tissue indicates that all indications excluding serum level of NO, iNOS havecorrelation relationship with activity of ulcerative colitis.Conclusion: 'kuijiefang' decoction can cure ulcerative colitis in rats models, and theeffect of this function is influenced by its dosage, namely there is a certain relationshipbetween its effect and dosage; In. the pathogenesis of ulcerative colitis, the abnormality ofoxygen metabolism and NO metabolism can be observed 'kuijiefang' decoction canregulate, correct these abnormal changes so as to make them return to or approach theirnormal conditions. And the level of MDA, SOD, NO, iNOS in colon tissue, the serumlevel of MDA, SOD have a dose relationship with activity of ulcerative colitis.Therefore, it can be concluded that the influences on NO and oxygen metabolism may bethe contributors for the effects of'kuijiefang' decoction in curing ulcerative colitis.
