Object: To observe the ratio of Th1/Th2 of rats model with endotoxemia prepared byendotoxin of lipopolysaccharide(LPS) injection at different time after LPSadministration, and conclude the relation of inflammatory and anti-inflammatoryresponse. To provide the data for the next study of intervention.Method: The method of endotoxin injection intravenously was applied to prepare themodel of endotoxemia in our study. Blood routine and biochemical variables weredetected. 60 healthy Wistar rats were randomly divided into 2 groups: control andmodel. Sera levels of IFN-γ, and IL-10 were detected at 0, 3h, 6h, 12h, 24h by ELISAin each group, and samples of lungs and spleens were observed.Results: The biochemical variables of model group increased significantly, the lesionsof lungs and spleens and the mortality were higher than the control. The blood level ofIFN-γ, gradually increased in general within 24h after the administration of LPS, andhad statistical difference at 24h point compared with the control (P<0.05) ; the bloodlevel of IL-10 had no significant difference compared with the control; the ratio ofIFN-γ/IL-10 increased in general, but had no statistical difference compared with thecontrol.Conclusions: The pro-inflammatory response, i.e. SIRS, dominated within 24 hoursafter the administration of endotoxin in model rats with endotoxemia. The evidence ofthe shift of Th1→Th2 hadn't been found in the study.
