Oviductus Ranae is a kind of animal tissue which is dried oviductus of Rana temporaria chensinensis. It is a famous traditional Chinese herbal medicines of rich nutrients and it is good for health especially for its functional factors. So it is widely used in food, medicine and livestock breeding industries. It has been reported that its active components(protein, fatey acid,steroid,inor-ganic compound and so on)is evidenced to have significant antitussive, expectorant effect,antiager,etc.Otherwise,there is no systematic study on the effect of Oviductus Ranae on Anti-fatigue function, according to these considerations,we take some researches as follows:1. In this paper,main technology of enzymatic hydrlysis of Forest frog to prepare low-molecular-weight peptide was studied.Functional evalution on antifatigue of the optimum OR compound was studied according to the test methods in"Functional assesment procedure and tests methods of health food"which is provided by MOH.The results show that: Enzymatic hydrocysates of Oviductus Ranae could obviously prolong the time of mouse loaded-swimming , significantly increase the liver glycogen, reduce the content of BUN,it indicates that Oviductus Ranae has a significant anti-fatigue function. 500mg/kg is the best dose.2. According to the index of the loaded-swimming time of mice ,the antifatigue function of 13 kinds of Chinese herb extracts add with OR was studied ,and four optimum Chinese herb extracts had been choosed from them.Then the OR compound compose of OR and the four optimum Chinese herb extracts was selected by using orthogonal design method.Finally,functional evalution on antifatigue of the optimum antifatigue OR compound was studied. The results show that: Oviductus Ranae compound with Rhodiola Rosea, Herba Cistanche ,Rhizoma Ginseng and Gynostemma Pentaphyllum can prolong the time of mouse loaded-swimming. The optimum OR compound obtained according to orthogonal design method could obviously prolong the time of mouse loaded-swimming , significantly increase the liver glycogen ,reduce the content of BUN and the blood lactic acid.These results indicated that the OR compound shown the effect of antifatigue.(2)The prescription composition of OR compound is consists of Oviductus Ranae,Rhodiola Rosea, Herba Cistanche ,Rhizoma Ginseng and Gynostemma Pentaphyllum ,which is 500 mg ,220 mg ,100 mg,200 mg and 80mg respectively. It was demonstrated that OR compound could obviously prolong the time of mouse loaded-swimming , significantly increase the liver glycogen, reduce the content of BUN and the blood lactic acid.3. SD rat model of swimming training was constructed and the expression ofα-actin of skeletal muscle was studied by the quantitative RT-PCR ( QRT-PCR )in order to investigate the molecular mechanism of the effect of Oviductus Ranae on antifatigue. The results indicated that:The OR compound also had a significant effect on the expression ofα-actin of skeletal muscle. It can enhance theα-actin expression of skeletal muscle in rats.4. Suitable for oral nanoemulsion of berberine was prepared by choosing appropriate of oil, surfactant, and co- surfactant. It was improved with new nanoemulsion carrier to the traditional preparations of berberine, by studying the pseudoternary phase diagrams.So it can be expected to provide the theory and practice for further development of Oviductus Ranae industry.The results show that : In the nanoemulsion of RH-60/Lecithin/IPM/1,3-butanediol system, nanoemulsion was very stable after three months storage. nanoemulsion in electron microscopy were small spherical drops with a mean diameter of 60 nm.In summury, Oviductus Ranae has a significant anti-fatigue function.The main approach of antifatigue is increase the liver glycogen, reduce the content of BUN and the blood lactic acid .the preparative nanoemulsion of Rhodiola Rosea and OR compound are good drug delivery system with stable quality and it is easy to prepare.
