The Expression Of IL-1α And The Effect Of Cyclosporin A In The EAM | | Posted on:2008-04-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Cao | Full Text:PDF | | GTID:2144360218451255 | Subject:Neurology | | Abstract/Summary: | PDF Full Text Request | | Objectives:1 To investigate the methods of inducing experimental autoimmune myositis(EAM) animal model.2 To investigate the effect of interleukin-1αin the attack of EAM rats.3 To assess the clinical effects of Cyclosporin in experimental autoimmune myositis rats and the changes of interleukin-1αin the model rats.Methods:1 Inducing experimental autoimmune myositis animal model with purified guinea pig muscle myosin(30mg/ml) mixed with CFA injected subcutaneously in SD rats 6 times.2 To treat EAM animals orally administered with Cyclosporin A 5mg/(kg·d), Prednisone 5mg/(kg·d) and then evaluate the effects of Cyclosporin according to the clinical symptoms and pathological examination.3 To detect the concentration of interleukin-1αin different group EAM animal models and evaluate the effects of interleukin-1αin the EAM animal models.Results:1 The clinical findings and pathologic changes in SD rats induced by partly purified muscle myosin of guinea pigs mixed with CFA injected subcutaneously were similar to the human's in idiopathic inflammatory myopathy; and the achievement ratio of the models induced by hypsiconcentration was higher. The changes in pathology were typical.2 Cyclosporin and prednisone were effective in the treatment of PM models according to the scores of clinical symptoms and pathology.3 There was significant up-regulation of the concentration of IL-1αin EAM rats. The t test showed the difference of the concentrations between the treatment groups (the Cyclosporin groups and the prednisone groups.) and control groups were significant statistically (P<0.05). There was no statistically significant difference in the Cyclosporin groups and the prednisone groups.Conclusions:1 The animal model induced by guinea pig myosin showed higher achievement ratio, and typical pathologic changes, so it was an ideal animal model to investigate PM.2 Cyclosporin was effective in the treatment of EAM.3 The upregulation of the expression of IL-1αmay be correlated with the onset of EAM.
